Background: Antigen specificity and IgG subclass could be significant in the natural history of Chagas' disease. The relationship between the different stages of human Chagas' disease and the profiles of total IgG and its subclasses were thus analysed here; they were directed against a crude T. cruzi extract and three recombinant antigens: the T. cruzi kinetoplastid membrane protein-11 (rKMP-11), an internal fragment of the T. cruzi HSP-70 protein 192-433, and the entire Trypanosoma rangeli HSP-70 protein.

Methods: Seventeen Brazilian acute chagasic patients, 50 Colombian chronic chagasic patients (21 indeterminate and 29 cardiopathic patients) and 30 healthy individuals were included. Total IgG and its subtypes directed against the above-mentioned recombinant antigens were determined by ELISA tests.

Results: The T. cruzi KMP-11 and T. rangeli HSP-70 recombinant proteins were able to distinguish both acute from chronic chagasic patients and infected people from healthy individuals. Specific antibodies to T. cruzi crude antigen in acute patients came from IgG3 and IgG4 subclasses whereas IgG1 and IgG3 were the prevalent isotypes in indeterminate and chronic chagasic patients. By contrast, the specific prominent antibodies in all disease stages against T. cruzi KMP-11 and T. rangeli HSP-70 recombinant antigens were the IgG1 subclass.

Conclusion: T. cruzi KMP-11 and the T. rangeli HSP-70 recombinant proteins may be explored together in the immunodiagnosis of Chagas' disease. Polarising the IgG1 subclass of the IgG response to T. cruzi KMP-11 and T. rangeli HSP-70 recombinant proteins could have important biological effects, taking into account that this is a complement fixing antibody.

Chinese hamster ovary (CHO) cells remain the most widely used host cell line for biotherapeutics production. Despite their widespread use, understanding endoplasmic reticulum (ER) stress conditions in recombinant protein production remains limited, often creating bottlenecks preventing improved production titers and product quality. Article Synopsis
  • - Enzymatic PET recycling is a promising eco-friendly alternative to mechanical recycling, but low production levels of PET hydrolases limit its large-scale use.
  • - Researchers improved FastPETase production in E. coli by co-expressing chaperones DnaK and DnaJ from Ideonella sakaiensis, achieving up to 2.5 times more soluble enzyme than existing chaperone methods.
  • - A combined strategy of chaperone co-expression and tagging the enzyme led to over 2 g/L of FastPETase in fermentation, with the crude product showing similar effectiveness in breaking down PET as the purified enzyme.

Key Laboratory of "Qin Medicine" Research and Development of Shaanxi Administration of Traditional Chinese Medicine, Shaanxi University of Chinese Medicine, Xianyang 712046, Shaanxi, China.

Article Synopsis
  • This study investigates the role of the heat shock protein gene Hsp70 in sclerotial formation and pathogenicity using techniques like RT-PCR and qRT-PCR to analyze gene expression at various growth stages and under stress conditions.
  • Results indicated that Hsp70 expression was highest in sclerotia compared to hyphae, significantly affected by cAMP stress and temperatures, showing the gene's importance in thermal stability.
  • Silencing Hsp70 resulted in a strain that could not form sclerotia, leading to decreased pathogenicity and resistance to fungicides, suggesting Hsp70 is crucial for stress resistance in the organism studied.

Article Synopsis
  • Research indicates that pathological forms of Tau are present in both Huntington's disease (HD) patients and animal models, suggesting HD may be seen as a secondary tauopathy alongside its main cause, a mutation in the huntingtin gene.
  • Experiments with zQ175 mice showed that injecting Tau fibrils led to cognitive and anxiety-like issues, as well as an increase in harmful mutant huntingtin aggregates in their brains.
  • Further studies with striatal cells revealed that Tau fibrils impair cell functionality and alter levels of heat shock proteins Hsp70 and Hsp90, indicating potential disruptions in protein quality control and suggesting a complex relationship between Tau and mutant huntingtin in HD.
