Introduction: In cancer, an extracellular and membrane bound localization of cathepsins contribute to the invasion of tumor cells at the basement membrane.

Methods: This is the first study that explored levels of cathepsins B (CatB), L (CatL) and their inhibitor cystatin C (CysC) in the cystic fluid (CF) of ovarian tumors (n = 110).

Results: CF contained considerable amounts of CatB, CatL and CysC. Remarkable differences in CatB and CatL and CysC CF levels were found between different histopathological tumor subtypes. Levels of CatB and CysC were significantly higher in CF of malignant serous tumors compared to those found in benign serous tumors (p = 0.010 and p = 0.001 respectively), whereas levels of CatL were significantly higher in CF of malignant mucinous tumors compared to those found in benign mucinous tumors (p = 0.035). CatB and CysC showed a strong correlation in the group of patients with malignant serous tumors (p < 0.001; R = 0.921) suggesting that the increase in CatB might be balanced by a corresponding increase in CysC.

Conclusion: Further studies are warranted to investigate cathepsins as possible prognostic biomarkers for the aggressiveness of ovarian cancer.

Article Synopsis
  • The study investigates the priming process of the Ebola virus (EBOV) glycoprotein GP involving furin and endosomal cathepsins but finds that furin cleavage isn't essential for the virus's ability to infect.
  • It demonstrates that different cell lines utilize various proteases for the processing of EBOV GP, highlighting inconsistencies in how the virus enters different types of cells.
  • The research reveals that an EBOV mutant lacking a furin cleavage site can still be activated by other proteases like TMPRSS2 and CatL, indicating the virus's adaptability and its dependency on proteolytic activation for entry into host cells.

Cathepsin B (CatB) is thought to be essential for the induction of lipopolysaccharide ( LPS)-induced Alzheimer's disease-like pathologies in mice, including interleukin-1β (IL-1β) production and cognitive decline. However, little is known about the role of CatB in virulence factor-induced IL-1β production by microglia. We first subjected IL-1β-luciferase reporter BV-2 microglia to inhibitors of Toll-like receptors (TLRs), IκB kinase, and the NLRP3 inflammasome following stimulation with LPS and outer membrane vesicles (OMVs).

The cysteine protease cathepsin S (CatS) is overexpressed in many tumors. It is known to be involved in tumor progression as well as antigen processing in antigen-presenting cells (APC). Recent evidence suggests that silencing CatS improves the anti-tumor immune response in several cancers.

Article Synopsis
  • - The study investigates how certain flavonols, specifically robinin (ROB) and gossypetin (GOS), interact with key targets related to COVID-19, including the virus's receptor binding domain and important proteases, showing their potential efficacy against the virus.
  • - ROB and GOS displayed strong binding affinities and stability in simulations, with lower binding free energy values indicating better interaction with the virus's targets; however, they do have some violations of Lipinski's rule regarding druglikeness.
  • - Although both compounds showed no significant toxicity and demonstrated potential therapeutic benefits for COVID-19, further testing is needed to assess their effects on liver enzymes and cellular metabolism before they can be considered for treatment.
