Background: Pseudomonas aeruginosa is an opportunistic pathogen that causes disease in immunocompromised individuals, burn victims, and cystic fibrosis patients. Strains that secrete ExoU induce host cell lysis and damage epithelial tissue, which can lead to severe outcomes including sepsis and mortality. ExoU is classified as an A2 phospholipase (PLA(2)) and activity is dependent on the eukaryotic protein, superoxide dismutase 1 (SOD1).

Methods: A sensitive and low background in vitro fluorescence-based assay was developed to detect ExoU activity using the fluorogenic substrate, PED6.

Results: The optimized assay enabled us to perform the first kinetic evaluation of the activation of ExoU (apparent K(m) of 13.2+/-1.5mumol/l PED6 and an apparent V(max) of 42nmol/min/mg). An inhibitor study using the inhibitor, methyl arachidonyl fluorophosphonate (MAFP), yielded an IC(50) of 13.8+/-1.1nmol/l and validated the use of high-throughput inhibitor screens using the assay. Most notably, the in vitro fluorescence-based activity assay was sensitive enough to detect catalytically active ExoU injected into eukaryotic cells.

Discussion: The use of the fluorescence-based activity assay to study the mechanism of ExoU activation may lead to the development of potential therapeutics to reduce P. aeruginosa-associated mortality.

Ethnopharmacological Relevance: The Chinese medicine sappanwood is primarily sourced from the dried heartwood of the medicinal plant Caesalpinia sappan Linn., which has been found with a variety of valuable properties including anti-inflammatory, anti-oxidant, and anti-viral effects. Preliminary investigations have demonstrated that sappanwood showed strong anti-SARS-CoV-2 M effects, but the key constituents responsible for SARS-CoV-2 M inhibition and their anti-M mechanisms have not been uncovered.

Lipid rafts are dynamic microdomains in the membrane, rich in cholesterol and sphingolipids, that are critical for biological processes like cell signalling, membrane trafficking, and protein organization. Their essential role is claimed in both physiological and pathological conditions, including cancer, neurodegenerative diseases, and viral infections, making them a key area of research. Fluorescence-based approaches, including super-resolution fluorescence microscopy techniques, enable precise analysis of the organization, dynamics, and interactions of these microdomains, thanks also to the innovative design of appropriate fluorescent probes.

Biodiesel is renewable energy source an alternative to conventional fossil fuels. The primary concern lies in detecting alcohol content in biodiesel, which can either be intentionally added by adulterants or remain in trace amounts from the refining process of biodiesel synthesis. In order to regulate the quality of biodiesel production, it is crucial to develop an analytical method for monitoring alcohol content in biodiesel.

Diabetes remains one of the most prevalent chronic diseases globally, significantly impacting mortality ratetables. The development of effective treatments for controlling glucose level in blood is critical to improve the quality of life of patients with diabetes. In this sense, smart optical sensors using hydrogels, responsive to external stimuli, have emerged as a revolutionary approach to diabetes care.

Background: Double-strand breaks (DSBs) are primarily repaired through non-homologous end joining (NHEJ) and homologous recombination (HR). Given that DSBs are highly cytotoxic, PARP inhibitors (PARPi), a prominent class of anticancer drugs, are designed to target tumors with HR deficiency (HRD), such as those harboring BRCA mutations. However, many tumor cells acquire resistance to PARPi, often by restoring HR in HRD cells through the inactivation of NHEJ.
