DNA synthesis of mammalian cells propagated in microplates can easily be measured if cell cultures incubated with [14C]thymidine are harvested on the glass fibre filters by a semiautomatic harvesting technique. Soon after infection with poliovirus, [14C]thymidine uptake of U cells (established, human amniotic cell line) is inhibited. This inhibition can be prevented by previous virus neutralization with antibody. Based on this effect a rapid, precise assay method was set up to determine neutralizing antibody titres against poliovirus. There was a good correlation between titres obtained by this assay and those obtained by 50% end point titrations in cytopathogenic effect inhibition assays.

Unlabelled: We evaluated 4'-[methyl-11C]thiothymidine ([methyl-11C]S-dThd) to obtain a thymidine analog that might prove simpler to use for imaging DNA synthesis and that might follow the same biochemistry as its surrogate.
Methods: [Methyl-14C]S-dThd was synthesized by rapid methylation of 5-trimethylstannyl-4'-thio-2'-deoxyuridine via a palladium-mediated Stille coupling reaction with 14C-methyl iodide. Degradation of [methyl-14C]S-dThd, when incubated in human blood, was analyzed by high-performance liquid chromatography (HPLC).

Zinc uptake is critical for cell proliferation. On the basis of the evidence that brain tumors are positively-imaged with 65Zn, cellular zinc uptake was studied under growth arrest and apoptosis to understand the relationship between cellular viability and zinc uptake. When NIH3T3 cells were cultured in albumin-coated dishes under the presence of serum, the viability of the cells detached from the extracellular matrix, which was determined with fluoresceine diacetate, was almost the same as the control cells cultured in untreated dishes.

Purpose: We determined whether changes in cellular reactive oxygen species correlated with mycobacteria internalization and bladder cancer cell death.
Materials And Methods: Reactive oxygen species and thiols in RT112 and MGH bladder cancer cells were determined using the fluorescence probes 5-(and 6)-carboxy-2', 7' dichlorodihydrofluorescein diacetate and monobromobimane. Superoxide and nitrite production were measured using bis-N-methylarcridinium nitrate and Griess reagents.
