The present review summarizes experimental data revealed while studying the mechanism of the adaptogenic effect of furostanol glycosides (FG) extracted from Dioscorea deltoidea Wall cell culture under the conditions of biotic stress in tomato plants Lycopersicon esculenium Mill. induced by the gall nematode Meloidogyne incognita Kofoid et White. Comparison of changes in isoprene content (phytosterines, tomatin, and carotenoids) and in the rate of oxidative processes in the leaves and roots of intact and treated plants evidence that FG cause nonspecific defense reactions resulting in the formation of systemic acquired resistance. This formation is presented by the enhancement in photosynthetic apparatus pigment fund, pigments of the violaxanthin cycle in particular, by activation of processes related to POL, and by increase in peroxidase activity-enzyme of antioxidant protection.

Article Synopsis
  • Yucca gloriosa leaves have a significant amount of steroid glycosides that undergo transformation from furostanol glycosides to spirostanol glycosides through the enzyme β-glucosidase.
  • There are two forms of β-glucosidase: Form I, which can hydrolyze both natural and synthetic substrates, and Form II, which only acts on synthetic substrates.
  • Form I shows a higher affinity for natural substrates, indicated by a lower Km value, and both enzyme forms operate best at a temperature of 37°C and a pH of 6.3-6.5.

Article Synopsis
  • Sawfly species specialized in feeding on Ranunculaceae plants can sequester specific furostanol saponins into their haemolymph, which may help them in defense against predators.
  • Research involving various chemical analyses and bioassays indicated that while one species predominantly fed on a non-nutritional stimulant from its host, both species showed differences in response to different plant fractions, with β-sitosterol identified as a nutritional stimulant.
  • The study found that the larvae of both sawfly species do not produce ecdysteroids endogenously but accumulate them from their diet, using them effectively as a deterrent against ant predators, especially in the initial days following simulated attacks.

Chemical investigations of higher plants, with particular attention paid to their steroidal glycosides, present a promising approach for generating anti-cancer agents from natural products. We conducted a systematic phytochemical investigation of nine higher plants-whole plants and rhizomes of Convallaria majalis, whole plants of Agave utahensis, roots of Adonis amurensis, seeds of Adonis aestivalis, bulbs of Bessera elegans, bulbs of Fritillaria meleagris, seeds of Digitalis purpurea, underground parts of Yucca glauca, and bulbs of Lilium pumilum-which led to the discovery of novel steroidal glycosides. The structures of these new constituents were determined based on spectroscopic data and chemical transformations.

Three undescribed compounds including two furosteroid glycosides (perfoloside and 22-O-methylperfoloside) and one stilbenedimer (perfolostilbene) together with 21 known compounds were isolated from the roots of Smilax perfoliata. The structural elucidation was established by extensive uses of HRMS, 1D and 2D spectroscopic techniques. The assignment of the stereocenters in perfolostilbene was based on NOESY data and ECD calculation.

Eight furostanol glycosides including five undescribed compounds, named tribufurostanosides A-E (1-5), and three known ones (6-8) were isolated from the fruits of Tribulus terrestris L. Their chemical structures were determined by the IR, HR-ESI-MS, 1D-, and 2D-NMR spectra. Furostanols 1-8 significantly inhibited nitric oxide production in LPS activated RAW 264.
