A microscale approach is described which screens conditions for recovering polyclonal antibodies from ovine sera by mixed-mode cation-exchange chromatography. The impact of pH and loading buffer salt concentration were assessed using robotically operated 20microL packed pipette tips. Low salt concentrations delivered capacities up to 41mg/mL, while only half this level was obtained at high salt concentrations. Two of the screened conditions were then tested in a 10mL packed bed and overall trends in capacity, yield and purity were found to be retained. Microscale pipette tips thus provided a useful basis for the rapid, approximate definition of a chromatography design space.

Extrusion-based printing of macroscopic architectures layer-by-layer offers new opportunities for constructing customized electromagnetic interference (EMI) shielding materials. However, current research primarily focuses on improving the printability of material inks by increasing contents and adding various modifiers, controllable construction of ultralight and robust macro-architectures with structural design at both macro- and micro-scales is still challenging. Herein, we develop a graphene oxide foaming ink enriched with air bubbles for direct-ink writing, enabling the creation of macroscopic graphene architectures with arbitrary geometries.

Fluorescent light-up aptamer/fluorogen pairs are powerful tools for tracking RNA in the cell, however limitations in thermostability and fluorescence intensity exist. Current in vitro selection techniques struggle to mimic complex intracellular environments, limiting in vivo biomolecule functionality. Taking inspiration from microenvironment-dependent RNA folding observed in cells and organelle-mimicking droplets, an efficient system is created that uses microscale heated water droplets to simulate intracellular conditions, effectively replicating the intracellular RNA folding landscape.
