Novel multitargeted antioxidants 3-6 were designed by combining the antioxidant features, namely, a benzoquinone fragment and a lipoyl function, of two multifunctional lead candidates. They were then evaluated to determine their profile against Alzheimer's disease. They showed antioxidant activity, improved following enzymatic reduction, in mitochondria and T67 cell line. They also displayed a balanced inhibitory profile against amyloid-beta aggregation and acetylcholinesterase, emerging as promising molecules for neuroprotectant lead discovery.

PD1-TLR10 fusion protein enhances the antitumor efficacy of CAR-T cells in colon cancer.

Int Immunopharmacol

January 2025

TriArm Therapeutics, Niudun Road 200, 201203 Shanghai, China. Electronic address:

Background: The immunosuppressive microenvironment negatively affects the efficacy of chimeric antigen receptor T (CAR-T) cells in solid tumors. Fusion protein that combining extracellular domain of inhibitory checkpoint protein and the cytoplasmic domain of stimulatory molecule may improve the efficacy of CAR-T cells by reversing the suppressive signals.

Methods: To generate optimal PD1-TLR10 fusion proteins, PD1 extracellular domain and TLR10 intracellular domain were connected by transmembrane domain from PD1, CD28, or TLR10, respectively.

Efficient oxygen evolution reaction (OER) catalysts with fast kinetics, high efficiency, and stability are essential for scalable green production of hydrogen. The rational design and fabrication of catalysts play a decisive role in their catalytic behavior. This work presents a high-entropy catalyst, FeCoNiCuMo-O, synthesized via carbothermal shock.

Non-precious transition metal-based electrocatalysts with high activities are promising candidates for substituting Pt- or Ru-based electrocatalysts in hydrogen evolution. In this study, we propose core-shell engineering to combine the amorphous NiCoP and crystalline CoP (a-NiCoP/CoP@NF), which requires an ultra-low overpotential of only 26 mV to achieve the benchmark current density of 10 mA cm. Furthermore, it achieves an industrial-level hydrogen evolution current density of 500 mA cm with excellent stability.

Oral Biomimetic Nanotherapeutics for Ulcerative Colitis Targeted Treatment by Repairing Intestinal Epithelial Barrier and Restoring Redox Homeostasis.

ACS Appl Mater Interfaces

January 2025

The structural disruption of intestinal barrier and excessive reactive oxygen/nitrogen species (RONS) generation are two intertwined factors that drive the occurrence and development of ulcerative colitis (UC). Synchronously restoring the intestinal barrier and mitigating excess RONS is a promising strategy for UC management, but its treatment outcomes are still hindered by low drug accumulation and retention in colonic lesions. Inspired by intestine colonizing bacterium, we developed a mucoadhesive probiotic -mimic entinostat-loaded hollow mesopores prussian blue (HMPB) nanotherapeutic (AM@HMPB@E) for UC-targeted therapy via repairing intestinal barrier and scavenging RONS.

Personalized Nanovaccine Based on STING-Activating Nanocarrier for Robust Cancer Immunotherapy.

ACS Nano

January 2025

Medical Research Center, The First Affiliated Hospital of Zhengzhou University, The Center of Infection and Immunity, Academy of Medical Sciences, Zhengzhou University, Zhengzhou, Henan 450001, China.

Tumor-specific T cells play a vital role in potent antitumor immunity. However, their efficacy is severely affected by the spatiotemporal orchestration of antigen-presentation as well as the innate immune response in dendritic cells (DCs). Herein, we develop a minimalist nanovaccine that exploits a dual immunofunctional polymeric nanoplatform (DIPNP) to encapsulate ovalbumin (OVA) via electrostatic interaction when the nanocarrier serves as both STING agonist and immune adjuvant in DCs.
