The pineal organ in fishes is a photoreceptive organ with dual outputs, neuroendocrine and neural. The neural projections of the zebrafish pineal were experimentally studied by means of tract-tracing with carbocyanine dyes (1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI), 3,3'-dioctadecyloxacarbocyanine perchlorate (DiO)). Double-labeling experiments were also performed in order to investigate the degree of overlapping of pineal, retinal or cerebellar projections in zebrafish. The pineal organ sends efferent fibers bilaterally to the rostral hypothalamus, thalamus, pretectum, posterior tubercle and the mesencephalic tegmentum. A few pinealofugal fibers could also be traced to the optic tectum. Most of the targets of the zebrafish pineal also receive retinal and/or cerebellar afferents, indicating a high degree of overlap with these projections. Since most of the targets of the pineal projections of zebrafish appear to be premotor and precerebellar centers, the neural output of the pineal organ is probably, because of its photoreceptive and circadian function, involved in photic and circadian modulation of these centers.

Monitoring the morphological and biochemical information of neurons and glial cells at high temporal resolution in three-dimensional (3D) volumes of in vivo is pivotal for understanding their structure and function, and quantifying the brain microenvironment. Conventional two-photon fluorescence lifetime volumetric imaging speed faces the acquisition speed challenges of slow serial focal tomographic scanning, complex post-processing procedures for lifetime images, and inherent trade-offs among contrast, signal-to-noise ratio, and speed. Located dorsally underneath a thin translucent skull in many teleosts, the pineal gland is a photoreceptive organ known as a key element of the circadian clock system. Nevertheless, the presence of additional routes of photoreception presents a challenge in determining its specific roles in regulating photic-related behavior. The study explored the ecotoxicological effects of chronic exposure to microplastic (MP) on adult zebrafish, focusing on environmentally relevant concentrations of polyethylene (PE) beads and polyester (PES). High-throughput untargeted metabolomics via UPLC-QToF-MS and 16S metagenomics for gut microbiota analysis were used to assess ecotoxicity in zebrafish exposed to varying concentrations of PE and PES. The VIP (Variable Importance in Projection) scores indicated PE exposure primarily impacted phospholipids, ceramides, and nucleotide-related compounds, while PES exposure led to alterations in lipid-related compounds, chitin, and amino acid derivatives.
