The psychopharmacological activity of a new compound--ONK-10--N-5(hydroxynicotinoyl)-L-glutamic acid was studied. It was shown in experiments on mice and rats that the compound possesses the pronounced antiamnestic and antihypoxic effects, does not disturb the conditioned reflex activity and the orientation behavior, has no anxiolytic activity and anticonvulsant properties, causes no disorder of movement coordination, is low toxic. ONK-10 is superior by its antiamnestic and antihypoxic effects to piracetam, meclophenoxat, demanol aceglumate and is not inferior to aniracetam.

Emopag, a new drug, preventively administered in doses of 10 and 30 mg/kg/day over 4 days produced a pronounced neuroprotective effect in the model of brain ischemia caused by gravitational overload and reduced animal mortality from 17 to 0%. The preparation more effectively corrected neurological deficit than the reference drugs Mexidol (in considerably larger doses of 30 and 90 mg/kg/day) and antihypoxic drug amtizol (30 mg/kg/day). Moreover, Emopag exhibited considerable antiamnestic activity comparable to that of Mexidol (in 3-fold higher doses); in a dose of 30 mg/kg/day Emopag was more effective than Mexidol and amtizol in the same dose.

Experiments with mice showed that in a multitude of acute hypoxia models (normobaric hypoxic hypoxia with hypercapnia, hypobaric, hemic and histotaxic) the antihypoxic action of a single intra-abdominal dose of melatonin surpasses greatly amtisol, the standard antihypoxic agent. Single melatonin injection produced a strong antiamnestic action on various amnestic models (scopolamine-induced, acute normobaric hypoxia with hypercapnia, and a combination of extreme factors) which was much better than of pyracetame, a well-known nootropic (mind-stimulating) drug. Increase of the melatonin dose from 1 mg/kg to 20 mg/kg amplified both the antihypoxic and antiamnestic effects.

The study of EEG and psychophysiological parameters and their correlations in 10 healthy volunteers after single oral administration of bromantan in comparison with placebo revealed favourable influence of the drug on operators' functional state. By EEG-profile (increase in middle-frequency alpha-rhythm power with lowering of powers in delta- and beta1-bands) bromantan is similar to many drugs with moderate psychostimulating, antiamnestic and antihypoxic properties. Bromantan does not alter the subjective state and principal components of operators' performance in untired men but promotes the appearance of EEG-signs of moderate vigilance rise and forming of productive action state coupled with favourable for precise motor performance tremor reduction.

2-(2-Aminoethoxy)-1-hydroxyethyl derivatives of bicyclic arenes (naphthalene, thianaphthene, benzofuran, and indole) were prepared and screened for antiamnestic (AA) and antihypoxic (AH) activities which were evaluated by measuring the reversing potency in electroconvulsion-induced amnesia and the protective effect against hypoxia, respectively, in mice. Compound 3o, 1-(benzo[b]thiophen-5-yl)-2- (2-diethylaminoethoxy)ethanol, showed the best AA and AH activity profile, being superior to our prototype compound, 2-(2-dimethylaminoethoxy)-1-phenylethanol (1). Elongation of the ethylene linkage in the side chain of 3o to 3- and 4-carbon moieties brought about a significant decrease in AH activity.
