Modified DNA containing silylated pyrene derivative at the 5'-terminus showed the obvious discrimination ability of the duplex formation, that the modified DNA showed the strong fluorescence emission in the presence of the complementary DNA, in spite of the very weak emission in the absence of the complementary DNA. The intensity of the fluorescence emission of the double strand found to increase about 10 times compared with that of the single strand. In this paper, we tried to disclose the detailed mechanism of such a discrimination ability of the silylated pyrene-modified DNA.

Evaluating tumor radiosensitivity is beneficial for the prediction of treatment efficacy, customization of treatment plans, and minimization of side effects. Tracking the mitochondrial DNA (mtDNA) repair process helps to assess tumor radiosensitivity as mtDNA repair determines the fate of the cell under radiation-induced mtDNA damage. However, current probes developed to monitor levels of DNA repair enzymes suffered from complex synthesis, uncontrollable preparation, limited tumor selectivity, and poor organelle-targeting ability.

Hard tick exoskeletons, composed primarily of chitin, pose a significant challenge for researchers attempting to extract genetic material. This study presents a simple modified, alternative method for extracting DNA from ethanol-preserved hard ticks. In this work, a new dual-signal fluorescence strategy based on nano-gold molecular beacon (MB) and in-situ generated silver nano-clusters (NCs) coupled with multiple amplification technique was developed for sensitive detection of miRNA (let-7b). miRNA can recognize both hairpin probe (HP) and auxiliary DNA, inducing dual-cycle amplification-process to release plenty of DNA S2. As the report probe carboxyfluorescein (FAM) was modified on Au nanoparticles (AuNPs), the fluorescent signal was quenched due to the fluorescence resonance energy transfer (FRET).
