We previously reported that a 3-pyridinecarbonitrile analog with a furan substituent at C-5 and a 4-methylindol-5-ylamino substituent at C-4, 1, was a potent inhibitor of PKCtheta (IC50=4.5 nM). Replacement of the C-5 furan ring of 1 with bicyclic heteroaryl rings, led to compounds with significantly improved potency against PKCtheta. Analog 6b with a 4-methylindol-5-ylamino group at C-4 and a 5-[(4-methylpiperazin-1-yl)methyl]-1-benzofuran-2-yl group at C-5 had an IC50 value of 0.28 nM for the inhibition of PKCtheta.

Comparison of the reactivity of sialyl chlorides and bromides based on -acetylneuraminic acid (Neu5Ac) and its deaminated analogue (KDN) in reactions with MeOH and -PrOH without a promoter revealed that the acetoxy group at C-5 in a molecule of a sialic acid glycosyl donor can destabilize the corresponding glycosyl cation making the S1-like reaction pathway unfavorable. A change to the S2-like reaction pathway ensures preferential formation of the α-glycoside.

Context: This article mainly studies three isomers of CHNO, namely 5-methyl-3,4-dinitro-1- (trinitromethyl) -1H pyrazole (1), 4-methyl-3,5-dinitro-1- (trinitromethyl) -1H pyrazole (2), and 3,5-bis (dinitromethyl) -4-nitro-1H-pyrazole (3). These three substances are excellent candidates for energetic materials, but their properties under external electric fields (EEF) have not been studied. Therefore, this article studied the properties of three isomers under EEF using density functional theory (DFT), and conducted statistical analysis on the obtained data, including the molecular structure, frontier molecular orbitals, surface electrostatic potential, and nitrate charge of the three isomers.
