FV100 is a promising new agent with extreme potency and specificity for varicella-zoster virus (VZV). It is the valyl ester pro-drug of Cf1743, the lead clinical candidate among the highly lipophilic bicyclic nucleoside analogue (BCNA) family discovered in Cardiff/Leuven. Cf1743 is unique amongst antivirals in terms of its structure and lipophilicity. It is exquisitely potent and selective for human VZV. FV100 has recently entered a randomized, controlled Phase II clinical trial for the treatment of shingles, sponsored by Inhibitex.

Autoimmune diseases are a heterogeneous group resulting from dysregulation or dysfunction of the immune system due to genetic predisposition and environmental triggers. It is common for these diseases to coexist. However, except for a single case secondary to varicella zoster virus infection, the association between juvenile idiopathic arthritis and acute cerebellitis has not been reported in the literature.

Herpes zoster (HZ) is a viral infection caused by the reactivation of endogenous and latent varicella-zoster virus that remains dormant in the cranial nerve or dorsal root ganglia. HZ occurs in a portion of the general population, with a higher incidence observed in high-risk individuals. Patients with impaired immunity, including human immunodeficiency virus infection, organ transplantation, old age, and cancer-related treatments such as chemotherapy (CT) and radiotherapy (RT) were found more prone to HZ infection.

An effective live zoster vaccine has been widely used around the world. Although no deaths occurred in the original large clinical trial, we analyzed 10 serious adverse events, including six deaths that have subsequently occurred in four countries. The goal is to define the viral pathogenesis of these unexpected adverse events secondary to a viremia with dissemination of the vaccine virus.
