Increasing legal requirements for risk assessment and efficacy testing in the dermo-cosmetic field have led to the development of alternative test methods. In this study, the porcine skin model was chosen to test the effect of irradiation on the penetration habits of UV filters and caffeine. For decades, the pig has been recognized as an experimental animal in biomedical research thanks to its morphological and physiological similarities to humans. In this study, we wanted to investigate the effect of UV irradiation on the absorption of octocrylene (OC) and benzophenone-3 (B3) sunscreens used under those circumstances and a model hydrophilic molecule, caffeine (Caf). These particular compounds were chosen due to their different lipophilic profiles. The percutaneous penetration of the two UV filters and Caf was studied after two simulated solar radiation doses of 61.4 kJ m(-2). After irradiation simulation, the total absorbed dose was increased for OC while for B3 and Caf it was lower. Thus, modifications in percutaneous absorption have been observed, and it appears that UV could play a crucial role in this process. Moreover, it has been observed that the lipophilic profile of the studied compounds affects percutaneous penetration when irradiated.

Continuous microenvironment modulation is an ongoing challenge in wound dressing, which includes excessive exudate absorption, oxygen delivery, bacterial inhibition and angiogenesis. Herein, we developed an construction strategy to fabricate a self-retaining double-layered wound dressing, where the top layer precursor was composed of Ca-containing polyvinyl butyral (PVB) solution dispersed with hydroxypropyl methylcellulose (HPMC) particles, and the bottom one consisted of sodium alginate (Alg) solution blended with Ag-doped mesoporous bioactive glass powders (Ag-MBG). When in use, both precursors were simultaneously squeezed out from the twin nozzles connected to the individual chambers of a twin-chambered syringe, whereby Ca in the top layer rapidly migrated downwards to crosslink Alg in the bottom layer, leading to the formation of an Alg/Ag-MBG (AA) functional hydrogel for filling an irregular wound.

This study aimed to profile metabolites from five Trichoderma strains and assess their cytotoxic and pharmacological activities, particularly targeting oral squamous cell carcinoma (OSCC). UHPLC-TOF-MS analysis revealed the presence of 25 compounds, including heptelidic acid, viridiol isomers, and sorbicillinol from the different Trichoderma extracts. Pharmacokinetic analysis showed moderate permeability and low interaction with P-glycoprotein, suggesting good drug absorption with minimal interference in cellular uptake.

Deep cutaneous wounds, which are difficult to heal and specifically occur on dynamic body surfaces, remain a substantial healthcare challenge in clinical practice because of multiple underlying factors, including excessive reactive oxygen species, potential bacterial infection, and extensive degradation of the extracellular matrix (ECM) which further leads to the progressive deterioration of the wound microenvironment. Any available individual wound therapy, such as antibiotic-loaded cotton gauze, cannot address all these issues. Engineering an advanced multifunctional wound dressing is the current need to promote the overall healing process of such wounds.
