One,1-dichloro-2,2 bis(p-chlorophenyl) ethylene (p,p'-DDE), the major metabolite of 2,2-bis(4-Chlorophenyl)-1,1,1-trichloroethane (DDT), is a known persistent organic pollutant and male reproductive toxicant. It has antiandrogenic effect. However, the mechanism by which p,p'-DDE exposure causes male reproductive toxicity remains unknown. In the present study, rat Sertoli cells were used to investigate the molecular mechanism involved in p,p'-DDE-induced toxicity in male reproductive system. The results indicated that p,p'-DDE exposure at over 30 muM showed the induction of apoptotic cell death. p,p'-DDE could induce increases in FasL mRNA and protein, which could be blocked by an antioxidant agent, N-acetyl-l-cysteine (NAC). In addition, caspase-3 and -8 were activated by p,p'-DDE treatment in these cells. The activation of NF-kappaB was enhanced with the increase of p,p'-DDE dose. Taken together, these results suggested that exposure to p,p'-DDE might induce apoptosis of rat Sertoli cells through a FasL-dependent pathway.

Cisplatin (CIS) is a widely used chemotherapeutic agent, but its side effects, such as oxidative stress, inflammation, and apoptosis, often lead to male reproductive damage. Oxidative stress, primarily caused by the excessive generation of reactive oxygen species (ROS), plays a critical role in disrupting testicular homeostasis, resulting in spermatogenic impairment and tissue injury. L-cysteine (CYS), a semi-essential amino acid with potent antioxidant and anti-inflammatory properties, may offer protection against CIS-induced oxidative damage.

Background: Periodontitis can induce systemic inflammation, and it may affect the testicles and male reproductive performance. This study investigated the effects of periodontitis on the testicles, reproductive performance, and offspring development in male rats.

Methods: Male Wistar rats were induced with periodontitis by ligating their first molars.

Background: Congenital hypothyroidism (CH) is a pathology that affects various organs, including the testicles. The mechanisms by which this condition alters fertility is unknown. This study aimed at determining if experimental CH affects gonocyte differentiation and arrests meiosis; and the possible role of the Sertoli cell (SC) in this condition.

Article Synopsis
  • The study investigates the harmful effects of di-(2-ethylhexyl) phthalate (DEHP) exposure on the testes of prepubertal rats, focusing on its impact on reproductive health.
  • DEHP exposure led to decreased testis weight, increased oxidative stress, structural disorganization of testes, and reduced numbers of spermatogenic and Sertoli cells, along with damaged mitochondria.
  • The research revealed that DEHP activates the TNF-α apoptosis pathway, resulting in impaired spermatogenesis and increased apoptosis, with similar effects observed using the active metabolite mono-(2-ethylhexyl) phthalate (MEHP) in vitro.

Background: Diabetes mellitus is a widely distributed endocrine disorder in the world. Altered reproductive function is a notable long-term consequence of type 1 diabetes mellitus (T1DM). In the current study, we assessed the effects of soya milk containing Lactobacillus casei and omega-3 on stereology of testes in type 1 diabetic rats.
