Article Abstract

Sporothrix globosa, reported from the USA, Europe, and Asia, is a recently described pathogenic species morphologically similar to Sporothrix schenckii. In this study, the phylogenetic affinities of 32 clinical and environmental isolates morphologically identified as S. schenckii, from Mexico, Guatemala, and Colombia, were assessed by cladistic analysis of partial sequences of the calmodulin gene using the maximum parsimony and neighbor-joining methods. The study revealed that one out of 25 isolates from Mexico (4%), one out of three isolates from Guatemala (33.3%), and two out of four isolates from Colombia (50%) belonged to S. globosa, while the other isolates belonged to S. schenckii sensu stricto. This is the first record of S. globosa from Mexico, and Central and South America.

Background: While metagenomic next-generation sequencing (mNGS) has been acknowledged as a valuable diagnostic tool for infections, its clinical validity and impact on patient management when using fresh tissue samples remains uncertain.

Methods: We conducted a retrospective cross-sectional study involving patients who underwent tissue mNGS at a tertiary hospital in China from February 2021 to February 2024, aiming to assess its ability to detect plausible pathogens and its clinical validity and impact.

Findings: A total of 520 mNGS results from 508 patients were analysed, detecting plausible pathogens in 302 (58.

Article Synopsis
  • Low-density neutrophils (LDNs) are a unique subset of neutrophils that play a role in systemic infections, but their specific function in subcutaneous fungal infections like sporotrichosis had not been studied until now.
  • This research found higher neutrophil counts in sporotrichosis patients, along with increased levels of granulocyte colony-stimulating factor (G-CSF), and a notable rise in CD16+CD66b+ LDNs compared to healthy individuals.
  • Additionally, LDNs showed impaired functions, with reduced ability to respond to infections, which correlates with disease severity in patients suffering from sporotrichosis.

Article Synopsis
  • In-58, a peptide derived from indolicidin, exhibits strong antibacterial effects with lower toxicity towards mammalian cells, unlike its precursor.
  • The peptide demonstrates significant antifungal activity against human pathogens, effectively inhibiting their growth without negatively interacting with existing antifungal medications.
  • Experimental results suggest that In-58 disrupts fungal cell wall integrity and DNA interaction, while also showing effectiveness in an infection model with low toxicity, indicating its potential use as a new treatment for sporotrichosis.

  • An 84-year-old Japanese farmer developed sporotrichosis after being scratched by his cat two months prior to seeking medical help.
  • A skin biopsy and tissue culture confirmed the diagnosis.
  • The patient was treated with oral itraconazole for 13 months, leading to resolution of his ulceration symptoms.

Article Synopsis
  • Sporotrichosis is a chronic fungal infection that often presents with atypical symptoms, making diagnosis challenging.
  • In a reported case, a 70-year-old man had a cutaneous lesion on his hand for six months, initially misdiagnosed as a bacterial infection, but further testing revealed it was sporotrichosis.
  • The patient was effectively treated with oral itraconazole, resulting in significant improvement of the lesion after two months.
