Background And Objective: Caveolin-1 is a marker protein of caveolae which is related with oncogenesis as a signal transduction hinge. This study was to investigate the effect of Caveolin-1 on the growth and apoptosis of doxorubicin-resistant human breast carcinoma cell line Hs578T/Dox.

Methods: Plasmids pCI-neo-caveolin-1 and pCI-neo-vector (control) were transfected into Hs578T/Dox cells, respectively. The expression of Caveolin-1 was detected by Western blot. Cell proliferation was detected by MTT assay. Cell cycle was detected by flow cytometry (FCM). Colony formation potential on soft agar was evaluated. Cell apoptosis was detected by FCM when cells were cultured for 48 h or cultured with staurosporine for 8 h.

Results: Caveolin-1 was overexpressed in Hs578T/Dox-cav-1 cells. The proliferation of Hs578T/Dox-cav-1 cells was obviously promoted when compared with that of Hs578T/Dox-vector cells (P<0.01). Colony size was larger in Hs578T/Dox-cav-1 group than in Hs578T/Dox-vector group. More colonies were formed in Hs578T/Dox-cav-1 group as compare with those in Hs578T/Dox-vector group (983.6+/-75.0 vs. 700.8+/-78.9, P<0.01). The proportions of cells at S and G2/M phases were higher in Hs578T/Dox-cav-1 group than in Hs578T/Dox-vector group. The proliferation rate of Hs578T/Dox-cav-1 cells was also higher than that of Hs578T/Dox-vector cells [(76.6+/-4.0)% vs. (58.0+/-4.1)%]. Over-expressed Caveolin-1 significantly reduced apoptosis index when the cells were cultured for 48 h [(5.7+/-0.5)% vs. (11.3+/-0.8)%] or cultured with staurosporine for 8 h [(13.8+/-1.2)% vs (21.4+/-1.9)%].

Conclusion: Overexpression of Caveolin-1 protein may promote the growth and anti-apoptosis ability of Hs578T/Dox cells.

Background: Bleomycin (BLM), an anticancer medication, can exacerbate pulmonary fibrosis by inducing oxidative stress and inflammation. Anti-inflammatory, anti-fibrotic, and antioxidant properties are exhibited by ganoderic acid A (GAA).

Aim: So, we aim to assess GAA's protective impact on lung fibrosis induced via BLM.

Microplastic (MP) represent a pervasive and escalating threat to aquatic ecosystems, impacting organisms from cellular to population levels. To investigate the immediate molecular impacts of MP exposure, we exposed Daphnia magna, a keystone species in freshwater ecosystems, to polystyrene microplastic particles (5 μm, 5 μg/L) for 48 h. Through proteomic and biochemical analyses, we identified extensive disruptions in key physiological pathways.

The Sprouty (SPRY) proteins are evolutionary conserved modulators of receptor tyrosine kinase (RTK) signaling. SPRY2 inhibits fibroblast growth factor (FGF) signaling, whereas it enhances epidermal growth factor (EGF) signaling through inhibition of EGF receptor (EGFR) endocytosis, ubiquitination, and degradation. In this study, we analyzed the effects of SPRY2 on endocytosis and degradation of FGF receptor 1 (FGFR1) using two human glioblastoma (GBM) cell lines with different endogenous SPRY2 levels.

Article Synopsis
  • Cinobufagin (CB) is a promising anticancer agent for lung cancer, showing potential through various preclinical studies that highlight its ability to inhibit tumor growth and promote cell death.
  • The review analyzed data from both lab-based (in vitro) and animal (in vivo) studies, finding that CB decreases cell viability while affecting key molecular pathways related to cancer.
  • Despite its effectiveness, limitations like the reliance on lab models, variability in study methodologies, and a lack of long-term safety data suggest more clinical trials are needed to confirm CB's safety and efficacy in humans.

Article Synopsis
  • Ouabain, a hormone, speeds up the progression of autosomal dominant polycystic kidney disease (ADPKD) by increasing cyst area and fibrosis specifically in ADPKD mice due to its interaction with Na,K-ATPase (NKA).
  • Researchers created a mouse model with a knockout of caveolin-1 (CAV1), the main structural protein of caveolae, to investigate the role of these structures in ouabain's effect on ADPKD.
  • The study found that without caveolae, the ADPKD mice did not show increased cyst progression or cellular changes in response to ouabain, indicating that caveolae play a crucial role in NKA signaling and the advancement of
