Novel proapoptotic Smac mimics/IAPs inhibitors have been designed, synthesized and characterized. Computational models and structural studies (crystallography, NMR) have elucidated the SAR of this class of inhibitors, and have permitted further optimization of their properties. In vitro characterization (XIAP BIR3 and linker-BIR2-BIR3 binding, cytotox assays, early ADMET profiling) of the compounds has been performed, identifying one lead for further in vitro and in vivo evaluation.

Hydrogen-bonded organic frameworks (HOFs) are under fast development in broad applications but have not been well explored for chemiresistive gas sensing yet primarily due to insufficient active sites. Herein, a new porphyrin-based HOF-199 is constructed by OH···O hydrogen bonds featuring layered networks and rich free oxygen (O) atoms, which is further exfoliated into few-layer nonosheets with more dangling O sites through an ultrasound-assisted liquid exfoliation method (namely L-HOF-199). Hypochlorous acid (HClO/ClO) is a common ROS that exhibits elevated activity levels in cancer cells. In this study, an ClO-triggered TADF probe, PTZ-MNI, was designed based on a naphthalimide core. PTZ-MNI self-assemble in aqueous environments, exhibiting significantly enhanced fluorescence that demonstrated typical aggregation-induced delayed fluorescence (AIDF) characteristics.

Near-infrared (NIR) triplet dyes are the cornerstones of cutting-edge biomedical and material applications. The difficulty in rational development of triplet dyes increases exponentially as the absorption wavelength shifts deeper into the NIR range. Although classical H-/J-typed packing of NIR dyes has the potential to enhance intersystem crossing (ISC) compared with that in single-chromophore dyes, the triplet state quantum yields remain limited in such strategy.
