Effects of 1,25(OH)2D3 on cardiovascular function.

1,25(OH)2D3 enhances myocardial contractility with pharmacological doses from 1 to 20 micrograms within 30-60 min. dp/dt modifications induced by 1,25(OH)2D3 are dose dependent and approach linearity from 1 to 10 micrograms. The inotropic effects are not dependent on extracellular Ca2+ levels; but cellular Ca2+ influx or internal Ca2+ movement may be implied. The effect seems not related to the described 1,25(OH)2D3 receptors which implies protein de novo synthesis. A modulation on other receptor-mediated substances acting on Ca2+ movements seems probable.

Ten stable, normocalcemic renal transplant patients with good allograft function, hyperparathyroidism, and variable hypophosphatemia were treated for 2 to 9 months with oral calcium carbonate and replacement doses of vitamin D analogues. Parathyroid hormone levels (PTH) and renal phosphate wasting were not autonomous or fixed but decreased with therapy. Although serum 1-25(OH)2D3 levels could be shown to rise appropriately during oral vitamin D therapy and fall afterwards, a separate study in a larger group of patients showed no effect of elevated parathyroid hormone or hypophosphatemia to increase endogenous 1-25(OH)2D3 levels.

A human cell line, designated as NISI, was established from a patient with erythroleukemia (FAB M6). The presence of a chromosome marker in NISI line indicates that it is derived from the leukemic clone which bears the common marker.
