This study demonstrates the feasibility of using Pseudomonas putida ONBA-17 to bioaugment a sequencing batch reactor (SBR) treating o-nitrobenzaldehyde (ONBA) synthetic wastewater. To monitor its survival, the strain was chromosomally marked with gfp gene. After a transient adaptation, almost 100% degradation of ONBA was obtained within 8 days as compared with 23.47% of the non-inoculated control. The bioaugmented reactor has a better chemical oxygen demand (COD) removal performance (96.28%) than that (79.26%) of the control. The bioaugmentation not only enhanced the removal capability of target compound, but shortened system start-up time. After the increase in ONBA load, performance fluctuation of two reactors was observed, and the final treating effects of them were comparable. What is more, denaturing gradient gel electrophoresis (DGGE) analysis of 16S rRNA genes via a combination of pattern comparison and sequence phylogenetic analysis was performed to uncover changes in sludge microbial communities. Only the members of alpha, beta and gamma subdivisions of Proteobacteria were identified. To isolate ONBA-degrading relevant microorganisms, spread plate was used and four bacterial strains were obtained. Subsequent systematic studies on these bacteria characterized their traits which to some extent explained why such bacteria could be kept in the system. This study will help future research in better understanding of the bioreactor bioaugmentation.

Background: Renal failure related death caused by diabetic kidney disease (DKD) is an inevitable outcome for most patients. This study aimed to identify the critical genes involved in the onset and progression of DKD and to explore potential therapeutic targets of DKD.
Methods: We conducted a batch of protein quantitative trait loci (pQTL) Mendelian randomization analysis to obtain a group of proteins with causal relationships with DKD and then identified key proteins through colocalization analysis to determine correlations between variant proteins and disease outcomes.

Flow chemistry has many advantages over batch synthesis of organic small-molecules in terms of environmental compatibility, safety and synthetic efficiency when scale-up is considered. Herein, we report the 10-step chemo-biocatalytic continuous flow asymmetric synthesis of cyproterone acetate (4) in which 10 transformations are combined into a telescoped flow linear sequence from commercially available 4-androstene-3, 17-dione (11). This integrated one-flow synthesis features an engineered 3-ketosteroid-Δ-dehydrogenase (ReM2)-catalyzed Δ-dehydrogenation to form the C1, C2-double bond of A ring, a substrate-controlled Co-catalyzed Mukaiyama hydration of 9 to forge the crucial chiral C17α-OH group of D ring with excellent stereoselectivity, and a rapid flow Corey-Chaykovsky cyclopropanation of 7 to build the cyclopropyl core of A ring.

Background: Wastewater systems are usually considered antibiotic resistance hubs connecting human society and the natural environment. Antibiotic usage can increase the abundance of both ARGs (antibiotic resistance genes) and MGEs (mobile gene elements). Understanding the transcriptomic profiles of ARGs and MGEs remains a major research goal.

Background: Ovarian cancers (OC) and cervical cancers (CC) have poor survival rates. Tumor-infiltrating lymphocytes (TILs) play a pivotal role in prognosis, but shared immune mechanisms remain elusive.
Methods: We integrated single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics (ST) to explore immune regulation in OC and CC, focusing on the PI3K/AKT pathway and FLT3 as key modulators.

The aim of this study was to investigate the loss of docosahexaenoic acid (DHA) from three supplements (two powders and one oil) after digestion (rumen and gastrointestinal) and their effects on the number and composition of rumen bacteria, using an in vitro approach. The concentration of supplements has a significant impact on the DHA loss rate and algal oil exhibited the highest rate of loss, but bioaccessibility was not significantly different from the other supplements. 16S rRNA sequencing showed that three DHA supplements altered the bacterial composition of in vitro batch cultures inoculated with rumen microorganisms from cows, and caused changes in the relative abundance of important bacterial phyla, families, and genera.
