Excessive auxin accumulation inhibits protocorm development during germination of Paphiopedilum spicerianum, delaying shoot meristem formation by downregulating boundary genes (CUC1, CUC2, CLV3) and promoting fungal colonization, essential for seedling establishment. Paphiopedilum, possess high horticultural and conservational value. Asymbiotic germination is a common propagation method, but high rates of protocorm developmental arrest hinder seedling establishment.

Deep learning has significantly accelerated drug discovery, with 'chemical language' processing (CLP) emerging as a prominent approach. CLP approaches learn from molecular string representations (, Simplified Molecular Input Line Entry Systems [SMILES] and Self-Referencing Embedded Strings [SELFIES]) with methods akin to natural language processing. Despite their growing importance, training predictive CLP models is far from trivial, as it involves many 'bells and whistles'.

Background: Rare diseases affect millions worldwide but sometimes face limited research focus individually due to low prevalence. Many rare diseases do not have specific International Classification of Diseases, Ninth Edition (ICD-9) and Tenth Edition (ICD-10), codes and therefore cannot be reliably extracted from granular fields like "Diagnosis" and "Problem List" entries, which complicates tasks that require identification of patients with these conditions, including clinical trial recruitment and research efforts. Recent advancements in large language models (LLMs) have shown promise in automating the extraction of medical information, offering the potential to improve medical research, diagnosis, and management.

Ebola and Marburg (EBOV and MARV) filoviral infections lead to fatal hemorrhagic fevers and have caused over 30 outbreaks in the last 50 years. Currently, there are no FDA-approved small molecule therapeutics for effectively treating filoviral diseases. To address this unmet medical need, we have conducted a systematic structural optimization of an early lead compound, -(4-(4-methylpiperidin-1-yl)-3-(trifluoromethyl)phenyl)-4-(morpholinomethyl)benzamide (), borne from our previously reported hit-to-lead effort.
