Background: Aberrant promoter methylation of Wnt inhibitory factor-1 (WIF-1) is a fundamental mechanism of epigenetic silencing in human cancers. Epigallocatechin-3-gallate (EGCG) has been reported to directly reactivate several methylation-silenced genes. The promoter demethylation and reactivation of WIF-1 has not previously been reported.

Materials And Methods: Methylation-specific PCR, sequencing analysis and RT-PCR analysis were performed to evaluate promoter demethylation of WIF-1 and WIF-1 expression, Western blot analysis and luciferase reporter assay were performed to evaluate expression of cytosolic beta-catenin protein and Tcf/Lef reporter activity.

Results: Promoter demethylation of WIF-1 and restoration of WIF-1 expression after EGCG treatment are demonstrated in H460 and A549 cell lines. EGCG also decreased cytosolic beta-catenin protein level and inhibited Tcf/Lef reporter activity.

Conclusion: These results suggest the potential therapeutic use of EGCG for the reversal of WIF-1 promoter methylation.

Delayed radiation-induced brain injury (RIBI) characterized by progressive cognitive decline significantly impacts patient outcomes after radiotherapy. The activation of NLRP3 inflammasome within microglia after brain radiation is involved in the progression of RIBI by mediating inflammatory responses. We have previously shown that sulfonylurea receptor 1-transient receptor potential M4 (SUR1-TRPM4) mediates microglial NLRP3-related inflammation following global brain ischemia.

Chemotherapy-induced neuropathic pain poses significant clinical challenges and severely impacts patient quality of life. Sodium ion channels are crucial in regulating neuronal excitability and pain. Our research indicates that the microRNA-30b (miR-30b) in rat dorsal root ganglia (DRG) contributes to chemotherapy-induced neuropathic pain by regulating the Nav1.

Arginine methylation is a common post-translational modification that plays critical roles in many biological processes. However, the existence of arginine demethylases that remove the modification has not been fully established. Here, we report that Myc-induced nuclear antigen 53 (Mina53), a member of the jumonji C (JmjC) protein family, is an arginine demethylase.

Article Synopsis
  • Diabetic wounds suffer from complications due to poor blood flow linked to vascular issues.
  • Previous attempts to use VEGF therapy to enhance healing haven't shown effective results, partly due to low levels of the enzyme phospholipase Cγ2 (PLCγ2).
  • By utilizing advanced gene-targeting techniques, specifically CRISPR/dCas9, researchers were able to demethylate the PLCγ2 gene in blood vessels, improving the effects of VEGF therapy and significantly enhancing blood flow and wound healing in diabetic patients.

Salt stress severely affects the growth and development of tomato. Strigolactones (SLs) and DNA methylation have been shown to be involved in the growth and development and response to salt stress in tomato. However, the regulation of SLs on DNA methylation in tomato under salt stress remains unclear.
