Zinc Lewis acid-catalyzed cyclization of ethenetricarboxylate derivatives 1 with 2-ethynylanilines has been examined. Reaction of 1,1-diethyl 2-tert-butyl ethenetricarboxylate 1b with 2-(trimethylsilylethynyl)aniline substrates in the presence of Zn(OTf)(2) gave bridged quinoline derivatives in 43-85% yield. The reaction of 1b with 2'-aminoacetophenone also gave the bridged quinoline derivative in 41% yield. Thermal reaction of bridged quinolines (180-190 degrees C) afforded indole derivatives in moderate to good yields.

These compounds were rationally designed considering pharmacophoric features and catalytic sites of the cross-talk proteins, synthesized, and assessed for their anticancer potential.

DNA gyrase is a bacterial type IIA topoisomerase that can create temporary double-stranded DNA breaks to regulate DNA topology and an archetypical target of antibiotics. The widely used quinolone class of drugs use a water-metal ion bridge in interacting with the GyrA subunit of DNA gyrase. Zoliflodacin sits in the same pocket as quinolones but interacts with the GyrB subunit and also stabilizes lethal double-stranded DNA breaks.

They might be derived from the corynantheine-type indole alkaloid via sequential oxidation and rearrangement.

Norfloxacin (NOR) is frequently detected in various water bodies and has the potential to promote the proliferation of NOR-resistant bacteria/genes in the environment. Dimeric Copper(I) Complex with a Disulfonamide-Bridged Core.

Pyridine-2-yl-sulfonyl-quinolin-8-yl-amide (psq) has produced the first sulfonamidato-bridged dicopper(I) complex, {Cu[κ-(μ-κ:κ-psq)]} containing the rhombic Cu(I)N core. The single crystal X-ray structure of this complex shows that two anionic psq ligands straddle the metal atoms via bridging sulfonamide N atoms to give a Cu···Cu distance of 2.9593(8) Å.
