Background: Deficits in interneuron and cholinergic circuits are noted in AD pathology, yet the precise mechanisms of their contribution to cognitive decline in the disease remain elusive. Neuronal Pentraxin 2 (NPTX2), a sensitive marker for synaptic activity and AD progression, is an immediate early gene expressed by pyramidal neurons that functions at excitatory synapses on Parvalbumin interneurons (PV-IN) to cluster AMPA receptors and strengthen circuit inhibition. NPTX2 is later shed from some synapses into the cerebrospinal fluid (CSF), where reduced NPTX2 levels inversely correlate with hippocampal volume and cognitive performance in individuals with AD/MCI.

Effective delivery of therapeutic agents for solid tumour treatment is impeded by multiple obstacles, such as aberrant interstitial fluid pressure and high density of the extracellular matrix, which causes impaired penetration to deep avascular tumour tissue that exists in a hypoxic immune cold environment. Only limited tumoricidal effects have been achieved with traditional nanomedicine due to its inefficient penetration and the multiple resistant effects that exist in the tumour microenvironment. Herein, a new chemo-dynamic immunotherapy (CDIT) is proposed based on a transcytosis tumour oxygenator (MnP) with effective chemo-dynamic effects.

Background: Temozolomide (TMZ) treatment has demonstrated, but variable, impact on glioma prognosis. This study examines associations of survival with DNA repair gene germline polymorphisms among glioma patients who did and did not have TMZ treatment. Identifying genetic markers which sensitize tumor cells to TMZ could personalize therapy and improve outcomes.

However, the deviation between the working and optimal pHs of nanozymes has been limiting their practical applications. Here we develop a strategy to modulate the microenvironmental pHs of metal-organic framework (MOF) nanozymes by confining polyacids or polybases (serving as Brønsted acids or bases).
