Addition of low levels of solid tris-(2-carboxyethyl)-phosphine hydrochloride to Applied Biosystems' R4A (2 mM) and R5 (0.17 mM) resulted in a coupled, phosphinedithiothreitol reducing system. Coupling the two reductants eliminated or greatly decreased interference from the oxidation product of dithiothreitol and allowed quantitation of phenylthiohydantoin (PTH)-Asp below the 400-fmol level. Use of the system resulted in a measurable increase in repetitive yield for beta-lactoglobulin, and instrument PTH standards were stable for more than 3 months.

Background: Myelin-laden foamy macrophages accumulate extensively in the lesion epicenter, exhibiting characteristics of autophagolysosomal dysfunction, which leads to prolonged inflammatory responses after spinal cord injury (SCI). Trehalose, known for its neuroprotective properties as an autophagy inducer, has yet to be fully explored for its potential to mitigate foamy macrophage formation and exert therapeutic effects in the context of SCI.
Results: We observed that trehalose significantly enhances macrophage phagocytosis and clearance of myelin in a dose-dependent manner in vitro.

True total-body and extended axial field-of-view (AFOV) PET/CT with 1m or more of body coverage are now commercially available and dramatically increase system sensitivity over conventional AFOV PET/CT. The Siemens Biograph Vision Quadra (Quadra), with an AFOV of 106cm, potentially allows use of significantly lower administered radiopharmaceuticals as well as reduced scan times. The aim of this study was to optimise acquisition protocols for routine clinical imaging with FDG on the Quadra the prioritisation of reduced activity given physical infrastructure constraints in our facility.
