Background: Methylotrophy describes the ability of organisms to grow on reduced organic compounds without carbon-carbon bonds. The genomes of two pink-pigmented facultative methylotrophic bacteria of the Alpha-proteobacterial genus Methylobacterium, the reference species Methylobacterium extorquens strain AM1 and the dichloromethane-degrading strain DM4, were compared.

Methodology/principal Findings: The 6.88 Mb genome of strain AM1 comprises a 5.51 Mb chromosome, a 1.26 Mb megaplasmid and three plasmids, while the 6.12 Mb genome of strain DM4 features a 5.94 Mb chromosome and two plasmids. The chromosomes are highly syntenic and share a large majority of genes, while plasmids are mostly strain-specific, with the exception of a 130 kb region of the strain AM1 megaplasmid which is syntenic to a chromosomal region of strain DM4. Both genomes contain large sets of insertion elements, many of them strain-specific, suggesting an important potential for genomic plasticity. Most of the genomic determinants associated with methylotrophy are nearly identical, with two exceptions that illustrate the metabolic and genomic versatility of Methylobacterium. A 126 kb dichloromethane utilization (dcm) gene cluster is essential for the ability of strain DM4 to use DCM as the sole carbon and energy source for growth and is unique to strain DM4. The methylamine utilization (mau) gene cluster is only found in strain AM1, indicating that strain DM4 employs an alternative system for growth with methylamine. The dcm and mau clusters represent two of the chromosomal genomic islands (AM1: 28; DM4: 17) that were defined. The mau cluster is flanked by mobile elements, but the dcm cluster disrupts a gene annotated as chelatase and for which we propose the name "island integration determinant" (iid).

Conclusion/significance: These two genome sequences provide a platform for intra- and interspecies genomic comparisons in the genus Methylobacterium, and for investigations of the adaptive mechanisms which allow bacterial lineages to acquire methylotrophic lifestyles.

Ceratostigma willmottianum Stapf is a unique chalk gland (salt-excreting) plant from China, with a salt gland structure that excretes white crystals of calcium carbonate (CaCO), which has potential biomineralization and carbon sequestration functions. Due to the narrow distribution of wild germplasm resources, there is a lack of diversity of new varieties to satisfy commercial development and scientific exploration. Therefore, we used ethyl methanesulfonate (EMS) mutagenesis to obtain new dwarf mutant germplasm, and analysed it in terms of morphology, growth, photosynthesis, salt glands, and excretion traits.

A novel Gram-stain-positive, aerobic, catalase-positive, oxidase-negative, non-motile, and rod-shaped bacterium with ibuprofen-degrading capacity, designated DM4, was isolated from the sewage of a wastewater treatment plant (WWTP) in Guangzhou city, China. Strain DM4 grew optimally at 0% (w/v) NaCl, pH 5.0-7.

Combinatorial Biosynthesis of 3--Carbamoylmaytansinol by Rational Engineering of the Tailoring Steps of Ansamitocins.

ACS Synth Biol

State Laboratory of Microbial Technology, Institute of Microbial Technology, Shandong University, Qingdao 266237, China.

Currently, most maytansine-containing antibody-drug conjugates (ADCs) in clinical trials are prepared with DM1 or DM4, which in turn is synthesized mainly from ansamitocin P-3 (AP-3), a bacterial maytansinoid, isolated from . However, due to the high self-toxicity of AP-3 to , the yield of AP-3 has been difficult to improve. Herein, a new maytansinoid with much lower self-toxicity to , 3--carbamoylmaytansinol (CAM, ), was designed and generated by introducing the 3--carbamoyltransferase gene together with the -methyltransferase genes from exogenous maytansinoid gene clusters into the 3--acyltransferase gene () deleted mutant HGF052.

The impact of periplasmic localisation on the functioning of the XoxF protein was evaluated in the well-studied dichloromethane-utilising methylotroph Methylorubrum extorquens DM4, which harbors only one paralogue of the xoxF gene. It was found that the cytoplasmic targeting of XoxF by expression of the corresponding gene without the sequence encoding the N-terminal signal peptide does not impair the activation and lanthanide-dependent regulation of the MxaFI-methanol dehydrogenase genes. Analysis of the viability of ΔxoxF cells complemented with the full-length and truncated xoxF gene also showed that the expression of cytoplasmically targeted XoxF even increases the resistance to acids.

Dichloromethane (DCM, methylene chloride) is a toxic halogenated volatile organic compound massively used for industrial applications, and consequently often detected in the environment as a major pollutant. DCM biotransformation suggests a sustainable decontamination strategy of polluted sites. Among methylotrophic bacteria able to use DCM as a sole source of carbon and energy for growth, DM4 is a longstanding reference strain.
