For safer and effective immunocontraception, zona (ZP3) and spermatozoa specific (YLP(12)) peptides have been presented on virus-like particles (VLPs) derived from Johnson grass mosaic virus coat protein. Immunization of FvB/cJ female mice with VLPs presenting YLP(12)-ZP3 fusion peptide and a physical mixture of VLPs presenting either YLP(12) or ZP3 epitope led to generation of specific antibody responses and a significant reduction in litters born per mice (p<0.005). Significant curtailment of fertility was also observed in animals immunized with adjuvnated ZP3 and YLP(12) synthetic peptides. These results suggest that VLPs can be used to present gamete epitopes for immunocontraception.

There is an urgent need to develop a better method of contraception which is non-steroidal and reversible to control world population explosion and unintended pregnancies. Contraceptive vaccines (CV), especially targeting sperm-specific proteins, can provide an ideal contraceptive modality. Sperm-specific proteins can induce an immune response in women as well as men, thus can be used for CV development in both sexes.

Problem: Recent studies in several laboratories are focused on delineating sperm antigens that are relevant to fertility and examining involvement of antibodies to these antigens in human immunoinfertility. Our laboratory has characterized two such antigens, namely fertilization antigen (FA-1) and YLP12 dodecamer sequence that are involved in sperm-oocyte binding. Sperm proteins are being investigated for their applications in the development of contraceptive vaccines (CV) in several laboratories. In the present study, various synthetic peptides based upon four sperm proteins, namely Izumo, fertilization antigen-1 (FA-1), YLP(12), and SP56 that are involved in various steps of the fertilization cascade were examined for their immunocontraceptive effect. The synthetic peptides were conjugated to four carrier proteins namely keyhole limpet hemocyanin (KLH), bovine serum albumin (BSA), chicken gamma-globulin, and chicken ovalbumin.
