Borane-mediated reduction of aryl and alkyl ketones with alpha-aryl- and alpha-pyridyloxy groups affords beta-hydroxy ethers in high enantiomeric purity (up to 99% ee) and in good yield, using as catalyst 10 mol % of spiroborate ester 1 derived from (S)-diphenylprolinol. Representative beta-hydroxy ethers are successfully converted to beta-amino ethers, with minor epimerization, by phthalimide substitution under Mitsunobu's conditions followed by hydrazinolysis to obtain primary amino ethers or by imide reduction with borane to afford beta-2,3-dihydro-1H-isoindol ethers. Nonracemic Mexiletine and nAChR analogues with potential biological activity are also synthesized in excellent yield by mesylation of key beta-hydroxy pyridylethers and substitution with five-, six-, and seven-membered ring heterocyclic amines.

Beta-hydroxy ketones can be reduced using a sequence of ruthenium-catalyzed silyl etherification followed by tetrabutylammonium fluoride (TBAF) promoted intramolecular hydrosilylation. Switching from TBAF to tetrabutylammonium difluorotriphenylsilicate (TBAT), even without first forming the silyl ether, gave cyclic dioxasilinane products. These somewhat sensitive compounds could be isolated pure by column chromatography using florisil as the stationary phase.

Picrachinentins A-F (-, respectively), six novel cyclopeptide alkaloid-type burpitides (CPABs), were isolated and fully elucidated from the EtOH extract of the stems and leaves of . Structurally, compounds - have a 14-membered paracyclophane ring system that was closed through an ether bond between the β-hydroxy amino acid and tyrosine and modified with a 4,5-methylenedioxybenzoyloxy (MDBz, and ) or hexanoyl (Hexa, , , , and ) group at the N-terminus. Interestingly, this is the first report on the isolation and characterization of CPABs from plants of the Simaroubaceae family.

Comparing Surface and Bulk Curing Processes of an Epoxy Vitrimer.

We used atomic force microscopy-based infrared spectroscopy (AFM-IR) and nanomechanical mapping (AFM-NM) to image the surface of a vitrimer, specifically dicarboxylic acid-cured diglycidyl ether of bisphenol A (DGEBA), to assess the curing process of a surface layer and compared this to the process in the bulk. We identified the β-hydroxy esters with various functionalities that are the key to form the cross-links for a system, including difunctional DGEBA and carboxylic acids. The IR peaks of the carbonyl group in generated ester groups are distinguished clearly from those in acids, allowing us to quantitatively assess the curing process at the surface and in the bulk.

This work demonstrates the electrochemical construction of 2-methyl-1-aryloxy-3-(arylselanyl)propan-2-ol/2-hydroxy-2-methyl-3-(arylselanyl)propyl 2-(2-hydroxy-2-methyl-3-(arylselanyl)propoxy)benzoate starting from aryl allyl ethers/allyl benzoates and diaryl diselenides under additive-free electrochemical conditions. This environmentally friendly method was achieved through constant current electrolysis in an undivided cell setup under acid, oxidant, or catalyst-free conditions. Additionally, this technique enabled the synthesis of a variety of β-hydroxy selenides including late-stage functionalization of drug derivatives in good to exceptional yields across various substrates under mild reaction conditions.

The Mukaiyama aldol reaction is generally a Lewis-acid catalyzed cross-aldol reaction between an aldehyde or ketone and silyl enol ether.
