Exploration of structure-activity relationship determinants in analogue series.

A computational methodology is introduced to systematically organize compound analogue series according to substitution sites and identify combinations of sites that determine structure-activity relationships (SARs) and make large contributions to SAR discontinuity. These sites are prime targets for further chemical modification. The approach involves the analysis of substitution patterns in "combinatorial analogue graphs" (CAG) and the application of an SAR analysis function to evaluate contributions of variable R-groups. It is applicable to analogue series spanning different potency ranges, for example, analogues taken from lead optimization programs or screening data sets (where potency differences might be subtle). In addition to determining key substitution patterns that cause significant SAR discontinuity, CAG analysis also identifies "SAR holes", i.e., nonexplored combinations of substitution sites, and SAR regions that are under-sampled in analogue series.

Identification of N,N-dimethylpentylone (DMP) in counterfeit "Ecstasy" and "Molly" tablets poses risk to public health due to its adverse effects. Little information is available regarding the pharmacological activity or relevant blood or tissue concentrations of DMP, and even less is known about other structurally related beta-keto methylenedioxyamphetamine analogues on recreational drug markets, such as N-propyl butylone. Here, a novel toxicological assay utilizing liquid chromatography-tandem quadrupole mass spectrometry (LC-QQQ-MS) was developed and validated for the quantitation of DMP and five related synthetic cathinones (eutylone, pentylone, N-ethyl pentylone (NEP), N-propyl butylone, and N-cyclohexyl butylone), with chromatographic resolution from isomeric variants and quantitation performed by standard addition.

Screening of the ChemDiv molecular library in cholesterol media against Mycobacterium tuberculosis (Mtb) H37Rv strain identified a novel isoxazole thiophene hit as a putative Rv1625c/Cya activator with a promising in vitro activity and good pharmacokinetic properties. Twenty-nine analogs were synthesized to assess the structure-activity relationships (SAR) to further improve potency. The most notable analog was P15, which showed an intramacrophage EC = 1.

Jurassic Osmundaceous Landscapes in Patagonia: Exploring the Concept of Ecological Stasis in the Deseado Massif, Argentina.

Herein, we report the presence of a plant paleocommunity, dominated by ferns of the family Osmundaceae, structurally preserved from the only known Mesozoic, fossiliferous geothermal deposits, from the La Matilde Formation (Middle-Upper Jurassic) in the Deseado Massif of Southern Patagonia, Argentina. A total of 13 siliceous chert blocks sampled in an area of approximately 250 m, preserving a monotypic assemblage dominated by Osmundaceae embedded within its original swampy substrate, are documented. Additional Osmundaceae and fewer ferns and conifers are present in the stratigraphically continuous, adjacent chert levels.

Spectrum sensing is recognized as a viable strategy to alleviate the scarcity of spectrum resources and to optimize their usage. In this paper, considering the time-varying characteristics and the dependence on various timescales within a time series of samples composed of in-phase (I) and quadrature (Q) component signals, we propose a multi-scale time-correlated perceptual attention model named MSTC-PANet. The model consists of multiple parallel temporal correlation perceptual attention (TCPA) modules, enabling us to extract features at different timescales and identify dependencies among features across various timescales.

A concise, transition metal-free four-step synthetic pathway has been developed for the synthesis of tetracyclic heterosteroidal compounds, 14-aza-12-oxasteroids, starting from readily available 2-naphthol analogues. After conversion of 2-naphthols to 2-naphthylamines by the Bucherer reaction, subsequent selective C-acetylation was achieved via the Sugasawa reaction and reduction of the acetyl group using borohydride, which resulted into the corresponding amino-alcohols. The naphthalene-based amino-alcohols underwent double dehydrations and double intramolecular cyclization with oxo-acids leading to one-pot formation of a C-N bond, a C-O bond and an amide bond in tandem, to generate two additional rings completing the steroidal framework.
