Published: June 1991

A lectin (LDL) has been isolated from carpophores of the edible mushroom, Lactarius deliciosus, using a combination of affinity chromatography on stromas of group O erythrocytes embedded in polyacrylamide gel and hydroxylapatite, and gel filtration chromatography. Its molecular weight, as determined by gel filtration, is about 37,000 and its structure is dimeric, with two distinct types of subunits (about 19,000 and 18,000). It appeared homogeneous on HPLC gel filtration, but exhibited microheterogeneity on isoelectric focusing. Amino acid analysis revealed that it contains a large amount of glycine. Hapten inhibition assaying indicated that the Lactarius lectin is most specific for D-Gal beta 1----3D-GalNAc. The lectin was found in the mycelium and its possible role in the fungus is discussed.

Azurin, a bacterial blue-copper protein, has garnered significant attention as a potential anticancer drug in recent years. Among twenty Pseudomonas aeruginosa isolates, we identified one isolate that demonstrated potent and remarkable azurin synthesis using the VITEK 2 system and 16S rRNA sequencing. The presence of the azurin gene was confirmed in the genomic DNA using specific oligonucleotide primers, and azurin expression was also detected in the synthesized cDNA, which revealed that the azurin expression is active.

A protein's molecular interactions and post-translational modifications (PTMs), such as phosphorylation, can be co-dependent and reciprocally co-regulate each other. Although this interplay is central for many biological processes, a systematic method to simultaneously study assembly states and PTMs from the same sample is critically missing. Here, we introduce SEC-MX (Size Exclusion Chromatography fractions MultipleXed), a global quantitative method combining Size Exclusion Chromatography and PTM-enrichment for simultaneous characterization of PTMs and assembly states.

Removal of dissolved organic matter in road runoff with sludge-based filters from the drinking water treatment plant.

Water Sci Technol

January 2025

China Construction Fifth Engineering Division Co., Ltd, Changsha, Hunan 410004, China.

Road runoff underwent treatment using a filter filled with sludge from drinking water treatment plants to assess its capacity for removing dissolved organic matter (DOM). This evaluation utilized resin fractionation, gel permeation chromatography, three-dimensional excitation-emission matrix fluorescence spectroscopy, and UV-Visible spectroscopy. The filter demonstrated enhanced efficiency in removing dissolved organic carbon, achieving removal rates between 70 and 80%.

Sterols and triterpene alcohols exist in free and esterified forms in edible oils. To date, only few studies have determined the content of free or esterified sterols and triterpene alcohols using gas chromatography-flame ionization detection (GC-FID). In this study, analytical conditions were optimized using free and esterified sterol standards.

DNAzyme approach for simultaneous mRNA cap and poly(A) tail length analysis: A one-step method to multiple quality attributes.

The dynamic landscape of mRNA technology highlights the need for innovative quality control (QC) strategies. In this study, we described an efficient one-step digestion approach for concurrent generation of 5'- and 3'-end fragments, enabling simultaneous mRNA capping and poly(A) tail analysis. Tailored 10-23-type DNAzymes, designed from 5'- and 3'-Untranslated Regions (UTRs), selectively cleaved mRNA to release both the 5'-Capped or uncapped short fragments and 3'-Poly(A) tail cleavage products.
