Background And Purpose: Monocytes-macrophages play a key role in the initiation and persistence of inflammatory reactions. Consequently, these cells represent an attractive therapeutic target for switching off overwhelming inflammatory responses. Non-steroidal anti-inflammatory drugs (NSAIDs) are among the most common drugs for the symptomatic treatment of rheumatic diseases. Their effects have been explained on the basis of cyclooxygenase (COX) inhibition. However, some of the actions of these drugs are not related to inhibition of prostaglandin synthesis.
Experimental Approach: We examined the effect of oxaprozin on apoptosis of immune complex-activated monocytes in comparison with drugs of the same class, and the signalling pathway that leads activated monocytes exposed to oxaprozin to apoptosis. In particular, we studied the activity of caspase-3, the involvement of IkappaB kinase (IKK)-nuclear factor kappaB (NF-kappaB) system and the activity of X-linked mammalian inhibitor of apoptosis protein (XIAP), Akt and mitogen-activated protein kinase (MAPK) in activated monocytes in the presence of oxaprozin.
Key Results: Immune complexes caused the inhibition of monocyte apoptosis. Oxaprozin reversed in a dose-dependent manner immune complex-induced survival of monocytes, without affecting the apoptosis of resting cells. Other NSAIDs are ineffective. The activity of oxaprozin was related to inhibition of Akt activation that, in turn, prevented p38 MAPK, IKK and NF-kappaB activation. Consistently, the inhibition of NF-kappaB activation reduced the production of the anti-apoptotic molecule XIAP, leading to uncontrolled activity of caspase 3.
Conclusions And Implications: These results suggest that oxaprozin exerts its anti-inflammatory activity also through COX-independent pathways. It is likely that oxaprozin-mediated inhibition of the Akt/IKK/NF-kappaB pathway contributes to its anti-inflammatory properties.

This study examined the energy-dependent physiological responses, including stress, innate immune, and antioxidant systems, as well as indicators of energy mobilization, in pacu (Piaractus mesopotamicus) exposed to intermittent cold, aiming to assess the correlations between these responses. The fish were acclimated to 28 °C, divided into two groups, a control group maintained at 28 °C, and another exposed to 16 °C for two 24 h periods with a 5-day interval between them. The fish were sampled at six time points: baseline (after acclimatization to 28 °C), 24 h after the 1st exposure to 16 °C, after 5 days of recovery at 28 °C, 24 h after the 2nd exposure to 16 °C, and after 24 and 48 h of recovery at 28 °C.

Background: We have recently shown that, during acute severe COVID-19, SARS-CoV-2 spike protein (S) induces a cascade of events resulting in T cell apoptosis. Indeed, by neutralizing the protease activity of its receptor, ACE2, S induces an increase in circulating Angiotensin II (AngII), resulting in monocytic release of reactive oxygen species (ROS) and programmed T cell death.

We evaluated both environmental (workplace) and biological (urine) exposure to the anesthetic isoflurane concerning the effects of such exposure on the hematological, hepatic, and renal parameters in veterinarians. Samples were collected from exposed and nonexposed (volunteers) groups to assess hematological and several biochemical markers, and isoflurane was measured in the workplace air and veterinarian's urine. Isoflurane was detected at high levels in the air and urine.

Sepsis is a risk factor associated with increasing neonatal morbidity and mortality, acute lung injury, and chronic lung disease. While stem cell therapy has shown promise in alleviating acute lung injury, its effects are primarily exerted through paracrine mechanisms rather than local engraftment. Accumulating evidence suggests that these paracrine effects are mediated by mesenchymal stem cell (MSC)-derived small extracellular vesicles (sEVs), which play a critical role in immune system modulation and tissue regeneration.
