Euscaphinin, a new ellagitannin dimer from Euscaphis japonica (THUNB.) KANITZ.

Published: April 2009

A new ellagitannin dimer named euscaphinin was isolated from the leaves of Euscaphis japonica (THUNB.) KANITZ. The structure of this dimer was determined on the basis of spectroscopic and chemical evidence. The resulting structure indicated that it was produced by intermolecular C-O oxidative coupling between the galloyl groups of molecules of 1(beta)-O-galloyl pedunculagin, the major ellagitannin of the leaves. The ellagitannins were not detected in the bark and wood part of the plant. This is the first example of the isolation of ellagitannins from Staphyleaceaeous plants.

Oenothein B (OeB), a dimeric ellagitannin with a macrocyclic structure, is reported to have beneficial effects, including antioxidant, antitumor, antiviral, and antimutagenic effects, on human health. Despite the remarkable properties of OeB, its role in neovascularization process has not yet been evaluated. Thus, this study aimed to evaluate the angiogenic activity of OeB using a chorioallantoic membrane (CAM) assay at different concentrations (6.

Tannins are a group of polyphenols that possess the ability to precipitate proteins, causing an undesirable astringent taste by interacting with salivary peptides. This interaction deactivates the digestive enzymes; therefore, tannins are considered as plant defense substances. The health benefits of tannins and related polyphenols in foods and beverages have been demonstrated by biological and epidemiological studies; however, their metabolism in living plants and the chemical changes observed during processing of foods and medicinal herbs raises some questions.

Despite the various biological activities exhibited by water chestnut (the fruit of the genus), the phenolic compounds present in its extract require comprehensive characterization. Accordingly, we analyzed a 80% methanol extract of commercially available water chestnut and identified a new hydrolyzable tannin dimer termed trapadin A. Additionally, 22 known compounds, including 10 hydrolyzable tannin monomers and 2 dimers, were also detected in the extract.

Oenothein B (OeB) is a dimeric ellagitannin with potent antioxidative, antitumor, immunomodulatory, and anti-inflammatory properties. Despite the promising activities of OeB, studies examining the genotoxic or protective effects of this ellagitannin on DNA are scarce. Therefore, to further comprehensively elucidate the chemopreventive profile of OeB, the aim of this study was to evaluate the mutagenic and antimutagenic actions of OeB using strains with the Ames test.
