Background: Pivotal Alzheimer's Disease (AD) trials typically require thousands of participants, resulting in long enrollment timelines and substantial costs. We leverage deep learning predictive models to create prognostic scores (forecasted control outcome) of trial participants and in combination with a linear statistical model to increase statistical power in randomized clinical trials (RCT). This is a straightforward extension of the traditional RCT analysis, allowing for ease of use in any clinical program.

Background: Dementia is strongly linked to increased care use, but the use of formal and informal care throughout dementia journey remains unclear.
Method: Within a population-based cohort study, we identified 240 older adults (aged ≥78 years) with who developed CIND and 155 with incident dementia. These participants were matched to 480 and 310 cognitively intact participants, respectively, and their formal and informal care use and care hours were compared with a control groups before and after diagnosis of cognitive disorders.

Background: Long-term care (LTC) home residents may be isolated or lonely. Social connection is important for their physical, mental and cognitive health, quality of life and care. Background: Loneliness is linked with risk for cognitive decline and dementia among older adults, but the degree to which it predicts future risk is unclear. To investigate if loneliness acts as a predictor of cognitive decline, this study employed a measurement burst design using data from the Einstein Aging Study, where loneliness and cognition were repeatedly assessed daily, for several days, across several years. In this type of data, a major challenge to detecting subtle cognitive changes is the presence of retest/practice effects.

Background: The use of blood biomarkers predictive of the presence of amyloid Aβ and ApoE4 is an effective tool to facilitate a timely AD diagnosis. About a third of dementia cases can be attributed to potentially modifiable risk factors, and the incidence of the disease can be reduced by addressing these factors. Memory Workshops integrated with early detection and actions on risk factors is an operational strategy that is both therapeutic and preventive.
