Objective: To explore the effect and mechanism of Poly I:C in inducing growth inhibition and apoptosis of human hepatocellular carcinoma SMMC-7721 cells.

Methods: SMMC-7721 cells were treated with different doses of Poly I:C for 24, 48, and 72 h, and the cell growth inhibition rate was analyzed with CCK-8 assay. The cell cycle and the apoptosis were analyzed using flow cytometry with Annexin-V and PI staining, and quantitative RT-PCR analysis were used to detect the expression of TLR3, TRIF, and IFN-beta mRNA in cells.

Results: In the cells exposed to Poly I:C at low, moderate, and high doses, the inhibitory rates was the highest in high-dose Poly I:C group, and at a given Poly I:C dose, prolonged exposure resulted in significantly increased cell growth inhibition rate (P<0.05). Flow cytometry showed that Poly I:C induced cell apoptosis in a time- and dose-dependent manner and significantly increased the percentage of G1-phase cells as compared with that in the control group. The mRNA level of TLR3, TRIF, and IFN-beta were also increased following Poly I:C treatment in comparison with the control group.

Conclusion: Poly I:C can induce significant growth inhibition and apoptosis of SMMC-7721 cells in a dose- and time-dependent manner possibly by causing cell cycle arrest and TLR3 signaling pathway activation that leads to IFN-beta production and cell apoptosis.

Ischemic stroke is a major cause of adult disability. Early treatment with thrombolytics and/or thrombectomy can significantly improve outcomes; however, following these acute interventions, treatment is limited to rehabilitation therapies. Thus, the identification of therapeutic strategies that can help restore brain function in the post-acute phase remains a major challenge.

Unusual Iron-Independent Ferroptosis-like Cell Death Induced by Photoactivation of a Typical Iridium Complex for Hypoxia Photodynamic Therapy.

ACS Appl Mater Interfaces

Ferroptosis is a unique cell death mode that relies on iron and lipid peroxidation (LPO) and is extensively utilized to treat drug-resistant tumor. However, like the other antitumor model, requirement of oxygen limited its application in treating the malignant tumors in anaerobic environments, just as photodynamic therapy, a very promising anticancer therapy. Here, we show that an iridium(III) complex (Ir-dF), which was often used in proton-coupled electron transport (PCET) process, can induce efficient cell death upon photo irradiation, which can be effectively protected by the typical ferroptosis inhibitor Fer-1 but not by the classic iron chelating agents and ROS scavengers.

Antimicrobial membranes based on polycaprolactone:pectin blends reinforced with zeolite faujasite for cloxacillin-controlled release.

Discov Nano

January 2025

National Nanotechnology Laboratory for Agriculture (LNNA), Embrapa Instrumentação, 1452 XV de Novembro St., São Carlos, SP, 13560-970, Brazil.

Multifunctional membranes applied to biomedical materials become attractive to support the biological agents and increase their properties. In this study, biopolymeric fibers based on polycaprolactone (PCL) and pectin (PEC) were reinforced with faujasite zeolite (FAU) for cloxacillin antibiotic (CLX) loading. FAU with a high specific surface area (347 ± 8 m g), high crystallinity and particles with a diameter of up to 100 nm were produced under optimized synthesis conditions (100 °C/4 h).
