Feasibility of using gravimetry to measure excess adsorption effects of a binary solvent system in a reversed-phase high-performance liquid chromatography column.

A modified method for weighing HPLC columns filled with solvent is described. The method prevents the loss of traces of solvent from within the threads of the column. The method was tested by obtaining the weights of a C18 column filled with 10 different organic solvents, showing a standard deviation on the order of 0.1%. A plot of gross column weight versus solvent density showed excellent linearity. The method was then used to weigh a column filled with several acetonitrile-water mixtures. The gross column weights were lower than would have been predicted from the density of the acetonitrile-water mixtures. A likely explanation is the existence of an adsorbed acetonitrile-rich liquid on the surface of the C18 adsorbent, which caused the lower than expected weights due to the lower density of pure acetonitrile relative to the bulk mixtures. The volume of pure acetonitrile required for the observed weight discrepancy was calculated. Based on the surface area of the column adsorbent, values of micromoles acetonitrile per square meter of surface area were determined. The values showed reasonable agreement with values obtained from published adsorption isotherm studies. This suggests that pycnometry may be a useful technique for adsorption studies. The limitations of the technique are discussed.

In a recent paper, we introduced the concept of structured microgroove columns as a potential solution to obtain perfectly ordered particle beds for nano- and micro-LC. In this concept, spherical particles are either individually positioned (single layer column) or stacked (multilayer column) in a series of interconnected micromachined pockets. After introducing a suitable method to fill the columns efficiently in a one-particle-per-pocket mode, the present contribution addresses the issue of sealing the filled columns and reports on the first flow tests conducted in a microgroove column device.

It has been demonstrated that when a low-molecular-weight salt solution flows through a polyelectrolyte gel, an electromotive force is generated, and its polarity depends on the sign of the polyelectrolyte network's charge. A novel L-shaped concrete-filled steel tube (CFST) column is proposed in this study. A finite element model of the column is developed using ABAQUS software to analyze its load transfer mechanism and axial compressive behavior. The effects of factors such as the steel strength, steel tube thickness, support plate configuration, and perforation of the support plates on the compressive performance of the column are investigated.

Irrespective of the rapid development of AAV-based gene therapy, the production of clinical-grade vectors has a bottleneck resulting from product-related impurities such as empty and partially filled capsids, which lack a functional recombinant genome. In the current study, we applied the sequential affinity chromatography (AC)- and anion-exchange chromatography (AEX)-based method for purification of AAV9 vector harboring single-stranded genome encoding the fusion of firefly luciferase (fLuc)-yellow fluorescent protein (YFP) under chicken beta actin (CBA) promoter. We assessed the efficiency of two different pre-packed cross-linked sepharose and one monolithic AEX columns following AC purification to separate fully encapsulated with recombinant DNA AAV vectors from byproducts.
