The design, synthesis and properties of a new derivatizing reagent, 1,3,5,7-tetramethyl-8-phenyl-(4-iodoacetamido)difluoroboradiaza-s-indacene (TMPAB-I), for thiol groups are presented. Using the derivatization of TMPAB-I with thiols, a new high-performance liquid chromatographic method for measuring low-molecular-weight thiol-containing compounds, including coenzyme A (CoA), glutathione, N-acetylcysteine, cysteine, homocysteine (HCys) and 6-mercaptopurine has been developed. The reaction of TMPAB-I with thiols is specific, fast and stable for both TMPAB-I and the derivatives. A baseline separation of all the six derivatives is achieved by isocratic elution on reversed-phase column within 20 min with detection wavelengths of 500 and 510 nm for the excitation and emission, respectively, and the limits of detection (signal-to-noise ratio=3) are from 1.8 fmol (CoA) to 14.0 fmol (HCys), respectively, per 20 microL injection. The utility of the proposed method has been validated by measuring thiol-containing compounds in human plasma samples from healthy persons and patients with hypertension, with recoveries of 94.2-106.8%.

A novel monomer, 9-bis[4-(2-hydroxyethoxy)phenyl]fluorene di(mercaptopropionate), with a highly refractive index, purity, and excellent UV-curable properties, is synthesized through an optimized Fischer esterification process, reacting 9,9-bis[4-(2-hydroxyethoxy)phenyl]fluorene with 3-mercaptopropionic acid. The structural characterization of this monomer is performed using Fourier-transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, high-performance liquid chromatography, and liquid chromatography-mass spectrometry. The synthesis conditions are optimized using a design-of-experiments approach.

Traditional photopolymerizations generally require an initiator for initiating the polymerization while few cases have created degradable chemical bonds. Moreover, the migration instability and cytotoxicity of photo initiators are posing issues to human health and the environment. Chiral macrocycles have emerged as attractive media for chromatographic enantioseparation due to their excellent host-guest recognition properties. In this study, a new chiral stationary phase (CSP) based on 1,1'-binaphthyl chiral polyimine macrocycle (CPM) was reported. The CPM was synthesized by one-step aldehyde-amine condensation of (S)-2,2'-dihydroxy-[1,1'-binaphthalene]-3,3'-dicarboxaldehyde with 1,2-phenylenediamine and bonded on thiolated silica via the thiol-ene click reaction to afford the CSP.

The present work reports on the preparation, characterization, and evaluation of a set of novel triphenyl-modified silica-based stationary phases without and with embedded ion-exchange sites for mixed-mode liquid chromatography. The three synthesized triphenyl phases differed in additionally incorporated ion-exchange sites. In one embodiment, allyltriphenylsilane was bonded to thiol-modified silica by thiol-ene click reaction, leading to particles with no ion-exchange sites.
