Sensitive and fast methods for the profiling of biologically important molecules are highly demanded. Mucins are densely O-glycosylated glycoproteins found at mucosal surfaces and are of great medical interest. Here we describe sensitive methods for the analysis of O-glycans from mucins using gel electrophoresis, and chromatography by nanoLC on graphite columns and structural analysis by electrospray mass spectrometry on a linear trap mass spectrometer.

In this paper, compact terahertz (THz) metachips for hyperspectral screening and quantitative evaluation of human cancer cells is reported. This pixelated resonant metachips feature the resonance channel from 1 and 3 THz frequency with a record-high quality factor (up to 230). Through the interactions of various cancer cells of different concentrations, high-dimensional spectral signatures are obtained, which are further transformed into a spatial map for labelling and quantification purposes.

We report the development and performance of a novel genomics platform, TempO-LINC, for conducting high-throughput transcriptomic analysis on single cells and nuclei. TempO-LINC works by adding cell-identifying molecular barcodes onto highly selective and high-sensitivity gene expression probes within fixed cells, without having to first generate cDNA. Using an instrument-free combinatorial indexing approach, all probes within the same fixed cell receive an identical barcode, enabling the reconstruction of single-cell gene expression profiles across as few as several hundred cells and up to 100,000 + cells per sample.

Developing a fluorescence sensing platform for point-of-care detection of low abundance biomarkers is highly valuable for early diagnosis of disease. Herein, a biomimetic fluorescence-enhanced platform based on photonic crystals and DNAzyme walker was constructed and further applied to visualize and quantify the miRNA-21 in biological samples. The DNAzyme walker was orthogonally activated by the target miRNA-21, which enabled the unlocking of the DNAzyme walker strand and the subsequently repeated substrate cleavage, thus generating enhanced fluorescence signals.
