Automation of the resazurin reduction test using fluorometry of microtitration trays.

Microtitration plate technique and -fluorometry was applied to automate the resazurin reduction test for monitoring bacterial numbers in broth cultures and milk. The effect of resazurin and resorufin concentration, bacterial species, growth medium, pH, and redox potential on the fluorescence response was studied. The timing of the appearance of maximum fluorescence was directly related to the logarithm of the number of colony-forming units (log CFU). Fresh milk and heat-treated milk contain interfering redox systems. The technique based on microtitration plate fluorometry, when fully automated, seems to provide a high-capacity system for analyzing bacterial numbers in foodstuffs and other media.

The spontaneous adsorption of proteins onto nanoparticles, known as the protein corona, provides a unique perspective for designing protein-sensing biosensors. This study proposes a tailored protein corona method mediated by Tween-20 and develops a reverse-capture approach for protein quantification assays. The protein-coated microplate captures titanium dioxide nanosheets (TiO-NS) in a phosphate buffer containing Tween-20 and generates fluorescence signals via the photocatalytic reduction of resazurin to resorufin, thereby indicating the amount of protein.

Highly sensitive and selective imaging of human-borne volatile organic compounds (VOCs) enables an intuitive understanding of their concentrations and release sites. While multi-VOC imaging methods have the potential to facilitate step-by-step metabolic tracking and improve disease screening accuracy, no such system currently exists. In this study, we achieved simultaneous imaging of ethanol (EtOH) and acetaldehyde (AcH), the starting molecule and an intermediate metabolite of alcohol metabolism, using a multiwavelength VOC imaging system.
