Calcium/calmodulin-dependent protein kinase II δ (CaMKIIδ) has a pivotal role in cardiac signaling. Constitutive and deleterious CaMKII "autonomous" activation is induced by oxidative stress, and the previously reported mechanism involves oxidation of methionine residues in the regulatory domain. Here, we demonstrate that covalent oxidation leads to a disulfide bond with Cys273 in the regulatory domain causing autonomous activity.

N-nitrosodimethylamine (NDMA), a probable carcinogenic disinfection by-product, can be formed with high molar yields following chloramination of ranitidine (RNTD), a histamine H-receptor antagonist. Although RNTD and some of its transformation products (TPs) have been studied under chlorination and photo-irradiation, the relationship between RNTD TPs and NDMA formation potential (NDMA-FP) remaining after those processes is still unclear. This study investigated the effects of chlorination and/or photo-irradiation on NDMA-FP derived from RNTD, simulating an urban water environment receiving treated wastewater.

Formation of toxic N-nitrosodimethylamine (NDMA) by chloramination of ranitidine, a drug to block histamine, was still an ongoing issue and posed a risk to human health. In this study, the effect of ozonation prior to chloramination on NDMA formation and the transformation pathway were determined. Influencing factors, including ozone dosages, pH, hydroxyl radical scavenger, bromide, and NOM, were studied.

Hypochlorous acid (HOCl) is a potent oxidant produced by the enzyme myeloperoxidase, which is released by neutrophils under inflammatory conditions. Although important in the immune system, HOCl can also damage host tissue, which contributes to the development of disease. HOCl reacts readily with free amino groups to form N-chloramines, which also cause damage in vivo, owing to the extracellular release of myeloperoxidase and production of HOCl.
