Objective: To determine the optimal condition for labeling rat mesenchymal stem cells (MSCs) using the fluorescent dye CFSE and the maximum time length allowed by CFSE staining for MSC tracing in vitro.
Methods: Rat MSCs were labeled with CFSE at different concentrations (2.5, 5.0, 10.0, 20.0 and 40.0 micromol/L) for 1, 5 or 10 min. The transfection efficiency and fluorescence intensity in the cells were measured by flow cytometry and fluorescence microscope to determine the optimal condition for MSC labeling. Under the optimal condition, the effect of CFSE on the growth of MSCs was evaluated by MTT assay, and flow cytometry and fluorescence microscope were used to determine the maximum time length following CFSE labeling to allow MSC tracing.
Result And Conclusion: Staining with CFSE at 20.0 micromol/L for 5 min was optimal for labeling rat MSCs in vitro, which allowed detection of the MSCs as long as 21 days after the labeling without obviously affecting the cell growth (P>0.05).

Developing the mechanism for MOF formation is crucial for the rapid development of new materials. This work demonstrates that Deuterium-NMR spectroscopy is the optimal inter-laboratory methodology for understanding the in-situ kinetics of metal-organic framework (MOF) formation. This method is facile, affordable, and allows for the isolation and monitoring of individual reagents by using one deuterated component while the remaining components are protonated.

Mechanical properties of engineered connective tissues are critical for their success, yet modern sensors that measure physical qualities of tissues for quality control are invasive and destructive. The goal of this work was to develop a noncontact, nondestructive method to measure mechanical attributes of engineered skin substitutes during production without disturbing the sterile culture packaging. We optimized a digital holographic vibrometry (DHV) system to measure the mechanical behavior of Apligraf living cellular skin substitute through the clear packaging in multiple conditions: resting on solid agar as when the tissue is shipped, on liquid media in which it is grown, and freely suspended in air as occurs when the media is removed for feeding.

Background: Computed tomography (CT) scans are widely used for evaluating children with acute atraumatic altered mental status (AMS) despite concerns about radiation exposure and limited diagnostic yield. This study aims to assess the efficacy of CT scans in this population and provide evidence-based recommendations.
Methods: A systematic review was conducted according to PRISMA guidelines.

Chronic back pain is a global health problem with significant impacts on physical and mental health, work ability, and quality of life. Back pain has an increased risk of becoming chronic, especially in patients with other chronic conditions. Treatment primarily focuses on nonpharmacological approaches.

Cartilage injuries are extremely common in the general population, and conventional interventions have failed to produce optimal results. Tissue engineering (TE) technology has been developed to produce neocartilage for use in a variety of cartilage-related conditions. However, progress in the field of cartilage TE has historically been difficult due to the high functional demand and avascular nature of the tissue.
