3-Benzoyl-7-dimethylamino-4-hydroxycoumarin was synthesized and its UV-vis spectra in various mixtures of MeOH and CH(2)Cl(2) were measured. The diketone lactone exists mainly in the exocyclic-enol form in protic solvents and the endocyclic-enol form in nonpolar solvents. Acetylation, amination and methylation products of the diketone lactone were characterized by X-ray crystallography. The results indicated that acetylation of coumarin-based diketone lactones occurs exclusively at the 4-hydroxy group, but that amination takes place specifically at the C-3 carbonyl group. Among the prepared compounds, amination products were found to exhibit photochromic property and colorimetric ion-sensing ability, and methylation product has the potential to function as a primary amine protection group.

Here, we report the preparation of lactones via Ni-catalyzed alkene hydroxylarylation and sequential intramolecular lactonization with O as a green oxidant and oxygen source. The bulky 1,3-diketone ligand is crucial by enabling Ni-catalyzed hydroxylarylation of alkenes, providing numerous phthalide and furanone derivatives with high efficiency under mild conditions. The synthetic value of this methodology was further demonstrated by the efficient synthesis of typhaphthalide and a monoamine oxidase B inhibitor.

On the basis of steric hindrance, one carbonyl group in a diketone can be reduced in a regioselective manner, with high enantioselectivity. The methodology can be extended to ketones with varied length of hydrocarbon chain spacing, and the products can be converted by oxidation to hydroxy esters or lactones without loss of enantiopurity.

Article Synopsis
  • An iron-catalyzed method for breaking carbon-carbon bonds and introducing difluoroalkyl groups is described as cost-effective.
  • This approach uses alkoxyl radical precursors to create fluorinated diketones through ring-opening and allows for the construction of difluoroalkylated lactones.
  • Both processes are efficient, operating under mild conditions with a wide variety of substrates and good compatibility with different functional groups.

A novel three-component strategy has been developed for the synthesis of iminosugars in good to excellent yields. This is the first report on the Mannich type addition of cyclic 1,3-diketones to aza-acetal derived from hydroxy-γ-lactone and arylamine to produce a novel series of aza-sugars with high selectivity.
