Published: February 2009

Aim Of The Study: This study was aimed to characterize the absorption, distribution and metabolism of ginseng in human subjects using pharmacokinetic experiments based on the metabolism by microflora obtained from the feces.

Materials And Methods: We examined how intestinal microflora transforms ginsenoside-Rb1 (G-Rb1) and ginseng extracts into Compound k (C-K) using in vitro experiments with microbial flora obtained from the feces of 32 male subjects. Subsequently, Cmax, Tmax and the area under the blood concentration curve (AUC) for C-K in plasma were estimated by quantitative analysis using LC-MS/MS. The correlation between C-K transforming activity and the AUC of C-K in plasma and other properties were analyzed.

Results: C-K was absorbed into the blood 24h after oral administration of ginseng, with average values for Tmax, Cmax, and AUC as follows: 10.76+/-2.07 h, 27.89+/-24.46(ng/ml), and 221.98+/-221.42(microg h/ml), respectively. There was a correlation between the C-K transforming activity of G-Rb1 and the C-K transforming activity of ginseng extract by intestinal microflora (Spearman's correlation coefficient=0.402, p<0.05).

Conclusion: The absorption of the final metabolites ginseng is independent of the metabolite transforming activity of intestinal microflora, but the Tmax, Cmax and AUC of the transformed metabolites are dependent on the activity of each individual's microbial flora.

The pathogenesis of IgA nephropathy and implications for treatment.

Nat Rev Nephrol

January 2025

Mayer IgA Nephropathy Laboratories, Department of Cardiovascular Sciences, University of Leicester, Leicester, UK.

IgA nephropathy (IgAN) is a common form of primary glomerulonephritis and represents an important cause of chronic kidney disease globally, with observational studies indicating that most patients are at risk of developing kidney failure within their lifetime. Several research advances have provided insights into the underlying disease pathogenesis, framed by a multi-hit model whereby an increase in circulating IgA1 that lacks galactose from its hinge region - probably derived from the mucosal immune system - is followed by binding of specific IgG and IgA antibodies, generating immune complexes that deposit within the glomeruli, which triggers inflammation, complement activation and kidney damage. Although treatment options are currently limited, new therapies are rapidly emerging that target different pathways, cells and mediators involved in the disease pathogenesis, including B cell priming in the gut mucosa, the cytokines APRIL and BAFF, plasma cells, complement activation and endothelin pathway activation.

GSK3β Inhibitors Inhibit TGFβ Signaling in the Human Trabecular Meshwork.

Eugene & Marilyn Glick Eye Institute, Department of Ophthalmology, Indiana University School of Medicine, Indianapolis, Indiana, United States.

Purpose: Primary open-angle glaucoma (POAG) is a leading cause of blindness, and its primary risk factor is elevated intraocular pressure (IOP) due to pathologic changes in the trabecular meshwork (TM). We previously showed that there is a cross-inhibition between TGFβ and Wnt signaling pathways in the TM. In this study, we determined if activation of the Wnt signaling pathway using small-molecule Wnt activators can inhibit TGFβ2-induced TM changes and ocular hypertension (OHT).

To enhance international and joint research collaborations in prostate cancer research, data from different sources should use a common data model (CDM) that enables researchers to share their analysis scripts and merge results. The OMOP CDM maintained by OHDSI is such a data model developed for a federated data analysis with partners from different institutions that want to jointly investigate research questions using clinical care data. The German Cancer Society as the scientific lead of the Prostate Cancer Outcomes (PCO) study gathers data from prostate cancer care including routine oncological care data and survey data (incl.
