Expression of glucocorticoid receptor protein in the hippocampus and frontal cortex of male and female rats during the 1st, 2nd, and 3rd weeks of life was studied by Western blot hybridization. In the frontal cortex, the concentration of receptor protein progressively increased from the 1st to the 3rd week of life; in females, expression of 94-kDa protein significantly surpassed that in males during the 1st week of life. In the hippocampus, expression of 94-kDa and 82-kDa proteins during the 1st week of life was higher in males. Moreover, expression of the major glucocorticoid receptor isoform (94 kDa) in this structure remained unchanged in all periods of the study in males, whereas in females it was low over the first 2 weeks of life and increased by the 3rd week. Variations in the expression of glucocorticoid receptors in the hippocampus of male and female rats coincide with changes in plasma corticosterone concentration during the early postnatal ontogeny.

Glucocorticoid excess causes bone loss due to decreased bone formation and increased bone resorption; miR-433-3p is a miRNA that negatively regulates bone formation in male mice by targeting Runx2 as well as RNAs involved in Wnt, protein kinase A and endogenous glucocorticoid signaling. To examine the impact of miR-433-3p on glucocorticoid-mediated bone loss, transgenic mice expressing a miR-433-3p tough decoy inhibitor in the osteoblast lineage were administered prednisolone via slow-release pellets. Bone loss was greater in control mice treated with prednisolone compared with miR-433-3p tough decoy mice due to higher osteoclast activity in the controls.

Objectives: To explore the mechanism by which histone deacetylase 1 (HDAC1) regulates steroid-induced apoptosis of mouse osteocyte-like MLO-Y4 cells.
Methods: MLY-O4 cells were treated with 400 nmol/L trichostatin A (TSA) or 1 mmol/L dexamethasone for 24 h or transfected with a HDAC1-overexpressing vector prior to TSA or dexamethasone treatment. The changes in the expressions of HDAC1, SP1, cleaved caspase-3 and Bax, SP1 acetylation level, cell proliferation, and cell apoptosis were examined.
