ClassRHO: a platform for classification of bacterial rieske non-heme iron ring-hydroxylating oxygenases.

We have developed an easy-to-use multiplatform classification tool, ClassRHO, which facilitates classification and comparison of bacterial Rieske non-heme iron aromatic ring-hydroxylating oxygenases (RHOs). Visualization and analysis can be generated on-the-fly by entering or uploading RHO query sequences. Pre-computed classifications were implemented for 42 standard RHO sequences. These 42 RHO sequences can be flexibly selected based on user requests. ClassRHO provides users with many options to view and analyze RHO sequences.

The shortage of corneal donors and the limitations in tissue engineering grafts, such as biocompatibility and mechanical properties, pose significant challenges in corneal transplantation. Here, for the first time, we investigate the effect of Rho kinase inhibitor Y-27632 and a dual media culture approach, including proliferative media (M1) and stabilizing media (M2), on rabbit limbal epithelial stem cells (LESCs), aiming to explore the feasibility of constructing corneal cell sheets in vitro through auto-bioprinting and assessing their corneal wound healing capacity in vivo. Y-27632 has primarily demonstrated significantly enhanced LESCs growth, proliferation, and reduced apoptosis.

Skin fibrotic diseases are characterized by abnormal fibroblast function and excessive deposition of extracellular matrix. Our previous single-cell sequencing results identified an enriched fibroblast subcluster in skin fibrotic tissues that highly expresses the actin cross-linking cytoskeletal protein Transgelin (TAGLN), which bridges the mechanical environment of tissues and cellular metabolism. Therefore, we aimed to investigate the role of TAGLN in the pathogenesis of skin fibrosis.
