p38 MAP kinase inhibitors have attracted considerable interest as potential agents for the treatment of inflammatory diseases. Herein, we describe a concise and efficient synthesis of inhibitor 1 that is based on a phthalazine scaffold. Highlights of our approach include a practical synthesis of a 1,6-disubstituted phthalazine building block 24 as well as the one-pot formation of boronic acid 27. Significant synthetic work to understand the reactivity principles of the intermediates helped in selection of the final synthetic route. Subsequent optimization of the individual steps of the final sequence led to a practical synthesis of 1.

The pentafluorosulfanyl (SF-) group has been the subject of a surge of interest in the past decade, but there is still little practicality associated with its synthesis and installation. Herein is reported the first continuous flow synthesis of pentafluorosulfanyl chloride (SFCl), the most common reagent for the synthesis of SF-substituted compounds. The synthesis is based on inexpensive and easy-to-handle reagents: sulfur powder (S), trichloroisocyanuric acid (TCCA) and potassium fluoride (KF).

X-linked hypophosphataemia (XLH) is a rare metabolic bone disorder caused by pathogenic variants in the PHEX gene, which is predominantly expressed in osteoblasts, osteocytes and odontoblasts. XLH is characterized by increased synthesis of the bone-derived phosphaturic hormone fibroblast growth factor 23 (FGF23), which results in renal phosphate wasting with consecutive hypophosphataemia, rickets, osteomalacia, disproportionate short stature, oral manifestations, pseudofractures, craniosynostosis, enthesopathies and osteoarthritis. Patients with XLH should be provided with multidisciplinary care organized by a metabolic bone expert.

Samul-tang (SM) is a traditional multi-ingredient herbal formula that is widely used in clinical practice for treating female infertility. Despite its known therapeutic benefits, the complexity of its action mechanisms owing to the combination of multiple compounds has limited its research and integration into modern pharmacological science. To address this challenge, we identified 38 herbal compounds as the major ingredients in SM and generated their transcriptome data from aged-mice ovaries by administering these individual compounds.
