Most of the 10-substituted triazolylartemisinin synthesized via the Huisgen 1,3-dipolar cylcoaddition of diastereomeric 10-azidoartemisinin (5, 6, and 7) with various alkynes (a-h) exhibit strong growth inhibition activity, even at sub-micromolar concentrations, against various cancer cell lines such as DLD-1, U-87, Hela, SiHa, A172, and B16. In particular, 10b and 10f showed a highly strong cytotoxicity.

From Pseudocyclic to Macrocyclic Ionophores: Strategies toward the Synthesis of Cyclic Monensin Derivatives.

J Org Chem

Department of Medical Chemistry, Faculty of Chemistry, Adam Mickiewicz University, Uniwersytetu Poznańskiego 8, 61-614 Poznań, Poland.

There has been a long search for a simple preparation of new cyclic analogues of ionophore antibiotics. We report a simple and general synthesis of three new cyclic derivatives of polyether ionophore, monensin A (MON). The application of the Huisgen 1,3-dipolar cycloaddition of azides and terminal alkynes to macrocyclization results in a concise, synthetic route to monensin lacton or lactam in only 4 steps.

Direct Click Bonding of Dissimilar Solid Materials Based on the Catalyst-Free Huisgen 1,3-Dipolar Cycloaddition.

Macromol Rapid Commun

January 2025

Department of Chemical Science and Engineering, Graduate School of Engineering, Kobe University, 1-1 Rokkodai, Nada-ku, Kobe, 657-8501, Japan.

Here, "direct click bonding" of solid materials is proposed, which is the direct bonding of solid surfaces via the formation of covalent bonds without any adhesive. The present study shows that the Cu-free Huisgen 1,3-dipolar cycloaddition reaction proceeds between solid surfaces displaying cyclooctyne and azide groups, and it achieved the strong bonding of dissimilar solid materials as a macroscopic reaction. The bonding strength obtained is sufficiently high for practical use, and the strength can be controlled by the surface density of the cyclooctyne groups.

Novel α-mangostin derivatives as promising antiviral agents: Isolation, synthesis, and evaluation against chikungunya virus.

Eur J Med Chem

December 2024

Department of Natural Products and Medicinal Chemistry, CSIR-IICT Hyderabad, India; Academy of Scientific and Innovative Research (AcSIR), Ghaziabad, 201002, India. Electronic address:

Investigations into fruit and vegetable processing residues (FVPRs) offer huge opportunities to discover novel therapeutics against many diseases. In this study, detailed investigation of Garcinia mangostana fruit peel extract led to the isolation and identification of ten known compounds (1-10). Further, a new series of α-mangostin derived sulphonyl piperzines, aryl alkynes and 1,2,3-triazole derivatives were synthesized using Huisgen 1,3-dipolar cyclo-addition reaction ("click" chemistry).

Cyclic peptides have higher stability and better properties as therapeutic agents than their linear peptide analogues. Consequently, intramolecular click chemistry is becoming an increasingly popular method for the synthesis of cyclic peptides from their isomeric linear peptides. However, assessing the purity of these cyclic peptides by mass spectrometry is a significant challenge, as the linear and cyclic peptides have identical masses.

Here, we report an electrochemical approach to generate an aldoxime-substituted nitrile oxide via the activation of nitromethane. The Cu-catalyzed Huisgen reaction of this 1,3-dipole with alkynes enables successful preparation of 48 new isoxazole aldoximes, which are typically challenging to synthesize by other methods, in 52 to 97% yields with excellent regioselectivity and chemoselectivity in a single step. Moreover, 20 3,3'-bisisoxazoles are prepared from the isoxazole aldoxime products in good yields via a two-step sequence.
