Dicer is responsible for producing small interfering RNA and microRNA as an essential component of RNA interference. The character of these small RNAs is determined by the structure of Dicer protein, which contains RNA HELICs, PAZ, RNaseand dsRNA binding domain. Different organisms have different numbers of Dicers with distinct but redundant functions. Evolutionally, the amount of Dicer in organism decreases, but its functions become multiple and extremely important. Studies on structure and function of Dicer will improve our understanding of the mechanism, as well as the evolutional rule and character, of RNAi and relative pathways. Here, we review the structural and functional character of Dicer.

In RNA interference (RNAi), long double-stranded RNA is cleaved by the Dicer endonuclease into small interfering RNAs (siRNAs), which guide degradation of complementary RNAs. While RNAi mediates antiviral innate immunity in plants and many invertebrates, vertebrates have adopted a sequence-independent response and their Dicer produces siRNAs inefficiently because it is adapted to process small hairpin microRNA precursors in the gene-regulating microRNA pathway. Mammalian endogenous RNAi is thus a rudimentary pathway of unclear significance.

Background: We aimed to elucidate the potential correlation between single-nucleotide polymorphisms (SNPs) in miRNA machinery genes and colorectal cancer (CRC) risk in an Iranian cohort.
Methods: We conducted a robust case-control study involving 507 participants, which included 213 patients diagnosed with CRC and 294 healthy controls at Research Institute for Gastroenterology and Liver Diseases in Tehran Province, Iran in 2018. The study focused on genotyping four specific SNPs, (rs14035), (rs197412), (rs2740348), and (rs3742330), using advanced ARMS-PCR and Tetra-primer ARMS-PCR techniques.

MicroRNAs (miRNAs) play a significant role in tumor progression, and regulating miRNA expression with small molecules may offer a new approach to cancer therapy. Among them, miRNA-20b has been found to be dysregulated in several cancers, including nonsmall cell lung cancer (NSCLC). Herein, an in silico high-throughput computer screen was conducted to identify small molecules that downregulate miR-20b using the three-dimensional structure of the Dicer binding site on pre-miR-20b.
