For the first time, the existence of a substrate adduct of a nickel superoxide dismutase (NiSOD) model, based on the first nine residues from the N terminus of the active form of Streptomyces coelicolor NiSOD, has been proven and the adduct has been isolated. This adduct is based on the cyanide anion (CN(-)), as a substrate analogue of the superoxide anion (O(2)(*-)), and the nickel metallopeptide H-HCDLPCGVY-NH(2)-Ni. Spectroscopic studies, including IR, UV/Vis, and liquid- and solid-state NMR spectroscopy, show a single nickel-bound cyanide anion, which is embedded in the metallopeptide structure. This complex sheds new light on the question of whether the mode of action of the NiSOD enzyme is an inner- or outer-sphere mechanism. Whereas discussion was previously biased in favor of an outer-sphere electron-transfer mechanism due to the fact that binding of cyanide or azide moieties to the nickel active site had never been observed, our results are a clear indication in favor of the inner-sphere electron-transfer mechanism for the disproportionation of the O(2)(*-) ion, whereby the substrate is attached to the Ni atom in the active site of the NiSOD.

Objective: To evaluate the toxic effects of the agent Relatox on mature outbred rats and mice in an acute experiment in comparison with the registered analogue Dysport.
Material And Methods: Based on the aim of experiment, the acute toxic effects of Relatox and Dysport were assessed on two animal species: rats and mice at intraperitoneal and intramuscular administration at dose levels that made it possible to calculate the toxicological parameter values (initially 10-150 U/kg with subsequent usage of additional doses 20 U/kg to 300 U/kg depending on the agent and route of administration). The LD values and other acute toxic parameters were calculated using probit analysis.

In this article we consider the theoretical and methodological implications of Deleuzian fabulation for research on recovery from drugs and alcohol as an alternative way of making and doing methods in sociology. The article draws on data produced as part of an ongoing interdisciplinary research collaboration, begun in 2019, with the visual artist and filmmaker Melanie Manchot, social scientists Nicole Vitellone and Lena Theodoropoulou, and people in recovery from drugs and alcohol engaged in the production of Manchot's first feature film STEPHEN. This project attends to the methodological practice of filmmaking as a way of thinking with and alongside colleagues from divergent disciplines about the role of methods, concepts and practices for confronting and resisting processes of stigmatisation.

Background: High content imaging-based functional precision medicine approaches have been developed and successfully applied in the field of haemato-oncology. For rheumatoid arthritis (RA), treatment selection is still based on a trial-and-error principle, and biomarkers for patient stratification and drug response prediction are needed.
Methods: A high content, high throughput microscopy-based phenotyping pipeline for peripheral blood mononuclear cells (PBMCs) was developed, allowing for the quantification of cell type frequencies, cell type specific morphology and intercellular interactions from patients with RA (n = 65) and healthy controls (HC, n = 33).
