The simultaneous determination of the antitubercular drugs rifampicin, pyrazinamide, isoniazid and the acetylisoniazid metabolite has been accomplished by LC, using a C-18 analytical column. The assayed drugs are usually administered together in the treatment of tuberculosis. Creatinine was also included in the chromatographic determination, in order to establish the curve of excretion of the drugs in urine. The chromatographic method uses a gradient flow in three steps, in conjunction with a programmed diode array photometric detection. In a 0.02 M potassium dihydrogen phosphate pH 7.0 buffer, a 5% (v/v) content of methanol for 1 min, a 8% (v/v) content of methanol for 3.4 min, and a 75% (v/v) content of methanol for 4 min were used. At 4.5 min, the wavelength value of detection was changed from 254 to 475 nm. Creatinine, acetylisoniazid, isoniazid and pyrazinamide were eluted in the first 4.5 min and rifampicin before 8 min. The method has been satisfactorily applied to the determination of the drugs in urine samples and in pharmaceuticals. The proposed LC method is simple, and a short time, less than 8 min is necessary for compounds elution.

Imaging Single Particle Profiler to Study Nanoscale Bioparticles Using Conventional Confocal Microscopy.

Single particle profiling (SPP) is a unique methodology to study nanoscale bioparticles such as liposomes, lipid nanoparticles, extracellular vesicles, and lipoproteins in a single particle and high throughput manner. The initial version requires the single photon counting modules for data acquisition, which limits its adoptability. Here, we present imaging-based SPP (iSPP) that can be performed by imaging a spot over time in the common imaging mode with confocal detectors.

Article Synopsis
  • Pharmacokinetics studies how drugs move through the body, focusing on absorption, distribution, metabolism, and excretion, while pharmacodynamics examines the drug's effects on the body.
  • The combination of pharmacokinetic and pharmacodynamic studies helps correlate drug concentrations with their therapeutic effects, providing valuable data for regulatory purposes and establishing administration guidelines.
  • Key considerations for pharmacokinetic studies include subject characteristics, dosages, sample types, collection times, and analytical methods to ensure accurate assessment of drug effects and concentrations.

Acute kidney injury (AKI) in paediatric kidney transplant recipients is common. Infection including urinary tract infection (UTI) and rejection are the most common causes in children. Surgical complications often cause AKI early post-transplant, whereas BK polyomavirus nephropathy rarely occurs in the first month post-transplant.

Globally, drug-impaired driving fatalities now exceed those from drunk driving, urging the need for on-site and roadside detection methods. In this study, a photothermal desorption and reagent-assisted low-temperature plasma ionization miniature ion trap mass spectrometer (PDRA-LTP-ITMS) was developed for on-site detection of drug-impaired driving. The pseudomultiple reaction monitoring (MRM) in PDRA-LTP-ITMS enables continuous ion selection during ion introduction and improved sensitivity to nearly 3-fold compared with the conventional full scan mode.
