ATP Regeneration from Pyruvate in the PURE System.

ACS Synth Biol

January 2025

Centre for Engineering Biology, Institute of Quantitative Biology, Biochemistry and Biotechnology, School of Biological Sciences, University of Edinburgh, Edinburgh EH9 3FF, U.K.

The "Protein synthesis Using Recombinant Elements" ("PURE") system is a minimal biochemical system capable of carrying out cell-free protein synthesis using defined enzymatic components. This study extends PURE by integrating an ATP regeneration system based on pyruvate oxidase, acetate kinase, and catalase. The new pathway generates acetyl phosphate from pyruvate, phosphate, and oxygen, which is used to rephosphorylate ATP .

Functionalization of polymer nanoparticles (NPs) with targeting peptides is of interest for drug delivery applications to enhance tumor accumulation and penetration. Herein, we evaluated the feasibility of two different methods for the attachment of a tumor-penetrating peptide LinTT1 (AKRGARSTA) to poly(ethylene glycol)-block-poly(ε-caprolactone) (PCL-PEG) NPs: (1) "post-conjugation" onto pre-formed nanoparticles, and (2) "pre-conjugation", the synthesis and purification of peptide-polymer conjugates and subsequent nanoprecipitation of the conjugates diluted with non-functionalized polymers. Conjugation of the labelled peptide via maleimide-thiol chemistry was verified by gel permeation chromatography (GPC) and fluorescence measurements.

Ribosomes scanning from the mRNA 5' cap to the start codon may initiate at upstream open reading frames (uORFs), decreasing protein biosynthesis. Termination at a uORF can lead to re-initiation, where 40S subunits resume scanning and initiate another translation event downstream. The noncanonical translation factors MCTS1-DENR participate in re-initiation at specific uORFs, but knowledge of other trans-acting factors or uORF features influencing re-initiation is limited.

The use of anesthetic agents as pre-operatory treatment to pearl seeding surgery can be stressful to organisms and activate various physiological response mechanisms. This study evaluated some parameters of the systemic antioxidant and immune responses in red abalone (Haliotis rufescens) exposed to 0.25 mL L eugenol (EUB), 3.

A cell-free gene expression system for prototyping and gene expression analysis.

Appl Environ Microbiol

December 2024

Department of Microbiology, Harvard Medical School, Boston, Massachusetts, USA.

Article Synopsis
  • The study introduces a cell-free gene expression (CFE) system that allows for transcription and translation of a specific anaerobic bacterium in the presence of oxygen, simplifying gene expression analysis.
  • Through optimizing cell extract preparation, the researchers significantly increased protein yields, finding that linear DNA templates yield better results than circular plasmids.
  • The CFE system shows promise for prototyping and investigating genetic expressions related to toxin production, particularly in hypervirulent strains linked to antibiotic resistance.
