A pair of monozygotic twins concordant for juvenile Parkinsonism are described. These twin sisters have lived together until 18 years old. Twin A noted tremor in the right hand and right-sided stiffness and slowness at the age 20. Initially, a marked improvement was shown with L-dopa treatment. However, one year after the beginning of treatment, dopa-induced dyskinesia appeared. Twin B noted tremor and right-sided stiffness and basal ganglia calcification in both patients. These twin patients suggest that genetic factors may play an important role in the cause of juvenile Parkinsonism.

A slowing of saccadic eye movements is one of the key symptoms of progressive supranuclear palsy and therefore represents a core functional domain of the current diagnostic criteria. However, there is considerable overlap between the saccade velocities of healthy people and patients in early stages. Therefore, a highly specific discrimination between patients and controls based on eye velocity often results in a considerable loss of sensitivity.

Objective: The study aims to identify risk factors associated with cognitive impairment in Parkinson's disease and to develop a predictive model to facilitate early clinical detection, diagnosis, and management, thereby enhancing patient prognosis and quality of life.
Methods: A total of 351 PD patients were enrolled from the Department of Neurology at the First Hospital of Shanxi Medical University between January 2022 and December 2023. Cognitive function was evaluated using the Montreal Cognitive Assessment (MoCA) scale, and patients were subsequently categorized into cognitively normal (PD-NC) and cognitively impaired (PD-CI) groups.

Parkinson's disease is an adult neurodegenerative disorder whose prevalence continues to rise. There is no proven therapeutic strategy capable of interfering with its unfavourable evolutionary profile. Current treatments remain symptomatic.

Parkinson's disease (PD) is a fatal neurodegenerative disease for which there are no still effective treatments able to stop or slow down neurodegeneration. To date, pathological mutations in the leucine-rich repeat kinase 2 (LRRK2) gene have been identified as the major genetic cause of PD, although the molecular mechanism responsible for the loss of dopaminergic neurons is still cryptic. In this review, we explore the contribution of Drosophila models to the elucidation of LRRK2 function in different cellular pathways in either neurons or glial cells.
